强酸型离子交换纤维PVF-g-SO3H对碱性氨基酸的分离研究

     

摘要

研究强酸型阳离子交换纤维PVF-g-SO3H对碱性氨基酸L-组氨酸和L-赖氨酸的吸附分离性能,并与732号树脂进行比较.结果表明PVF-g-SO3H纤维不仅对单组份赖氨酸或组氨酸的吸附率比732号树脂高约1.5%~3.9%,而且其洗脱率高约12.5%~13.4%,同时对混合氨基酸的洗脱峰既强而且分辨率高.再生后纤维和树脂的总提取率都有微小的下降,但纤维的再生提取率仍高于732号树脂10%以上.实验的最佳洗脱条件为:洗脱液柠檬酸-柠檬酸钠的pH值为4.6,填充柱长径比为20,洗脱速率为1 mL/min.%he adsorption and separation properties of strong acidic ion exchange fiber PVF-g-SO3H for basic amino acids lysine and histidine were investigated by comparison with general used No.732 resin. The experimental results showed that not only the adsorption rates of PVF-g-SO3H fiber are 1.5%~3.9% higher,but also its elution rates are 12.5%~13.4% higher than No.732 resin for single component lysine or histidine. Moreover, the strength of elution peak and resolution of lysine/histidine mixture over PVF-g-SO3H fiber column were obvious higher than that over No.732 resin. Although the total extraction rates of both fiber and resin are slightly decreased after regeneration, the former still keeps over 10% higher elution rates than the latter. In this study, the optimum elution conditions were as follows: citric acid-triple sodium citric acid buffer with pH 4.6, length/diameter ratio of column 20 cm, elution rate 1 mL/min.
