高效液相色谱-蒸发光散射法测定马鞭草中齐墩果酸与熊果酸的含量

         

摘要

建立了高效液相色谱-蒸发光散射检测法测定马鞭草药材中齐墩果酸和熊果酸含量的方法,色谱柱为PinnacleⅡC18 (250mm× 4.6mm,5m),流动相为甲醇-0.2%冰醋酸(体积比82.5∶:17.5);淋洗速度1mL/min;柱温35℃;蒸发光散射检测器检测,漂移管温度为85℃;气体流速为2.2L/min.齐墩果酸在0.317~1.585μg范围内(r =0.9997),熊果酸在0.585~2.925 g范围内(r=0.9999)线性关系良好.药材中齐墩果酸和熊果酸的回收率(n=6)分别为100.4%和97.6%,标准偏差分别为1.84%和2.34%.所建方法准确,重现性好,可作为马鞭草药材的质量控制方法.%A method for the determination of oleanolic acid and ursolic acid in Verbena officinalis L by high performance liquid chromatography-evaperative light scattering detector (HPLC-ELSD) was established. A Pinnacle II C18 column (4. 6mmX250mm) was used for separation at 35℃. The mobile phase was metha-nol-0. 2% acetic acid (82. 5 :17. 5) with the flow rate of lmL/min. The drift tube temperature was 85℃ and the gas flow rate was 2. 2L/min. The oleanolic acid had a good linearity in the range of 0. 317 ~ 1. 585μg ( r =0. 997) and ursolic acid in the range of 0. 585~2. 925μg ( r = 0. 9999). The average recoveries of oleanolic acid and ursolic acid were 100. 4% (RSD=1. 84%) and 97. 6% (RSD=2. 34%), respectively. The method is accurate and repeatable and could be used for the quality control of Verbena officinalis L.
