新型吡唑并吡嗪酮衍生物ppo3a、ppo3b和ppo3i对HeLa细胞增殖和凋亡的影响

         

摘要

Objective To find out whether the novel pyrazolo[1,5-a]pyrazin-4(5H)-one derivatives ppo3a, ppo3b, and ppo3i can inhibit cervical cancer (HeLa cell) and to explore the inhibition mechanism. Methods The HeLa cells have been treated by different concentrations of ppo3a, ppo3b and ppo3i for 48 h. Then the proliferative activity was determined by the SRB (sulforhodamine B) assay, and the apoptosis was determined by Hoechst 33258 staining assay. Lactate dehydrogenase(LDH) was also tested. Results The proliferation of the HeLa cells were significantly suppressed by these compounds in a dose-dependent manner. However, there were no significant differences (P>0.05) in the LDH release of the cells between the control group and the compounds treatment groups. Significant apoptosis was induced in HeLa cells after incubated with 40 μM of ppo3a, ppo3b or ppo3i for 48 h. Conclusion The compounds ppo3a, ppo3b and ppo3i could inhibit the prolif-eration of HeLa cells by inducing cellular apoptosis. These compounds may be a clue to find effective and safe drug for cervical cancer.%目的:研究新型吡唑并吡嗪酮衍生物ppo3a、ppo3b和ppo3i对HeLa细胞增殖和凋亡的影响。方法采用不同浓度(20,40,80μM)的ppo3a、ppo3b、ppo3i处理Hela细胞48 h,通过SRB(sulforhodamine B)测定细胞的增殖活性,检测乳酸脱氢酶(LDH)活性,Hoechst 33258染色法检测细胞的凋亡情况。结果 ppo3a、ppo3b、ppo3i能以浓度依赖的方式抑制HeLa细胞的增殖,但对照组与处理组之间LDH活性比较均无显著性差异(P>0.05),而40μM ppo3a、ppo3b、ppo3i处理48 h能够显著诱导HeLa细胞凋亡。结论 ppo3a、ppo3b和ppo3i可显著抑制HeLa细胞增殖,这与其诱导细胞凋亡途径有关。
