地不容生物碱成分研究

         

摘要

To study the alkaloids of Stephania epigaea,silica gel and Sephadex LH-20 column chromatography were employed to separate alkaloids from the roots of this plant. HRESIMS,NMR,and specific rotatory were adopted to determine the structure of the pure compounds. Seven alkaloids including three morphine alkaloids,sinomenine(1),sinomenine N-oxide(2),and sinoactine(3), and four protoberberine alkaloids,tetrahydropalmatine(4),corydalmine(5),stepholidine(6),and isocorypalmine(7)were isolated and identified from S. epigaea. Compounds 2 and 7 were isolated from this plant for the first time. Compound 3 showed obvious ace-tylcholinesterase(AChE)inhibitory effect with the IC50 value of 3.03 μM.%为了研究地不容的生物碱成分,采用硅胶柱层析和Sephadex LH-20柱层析进行化合物的分离纯化,采用HRESIMS,NMR以及比旋光度测定等方法对化合物结构进行鉴定。从地不容中分离鉴定了7个化合物,包括3个吗啡型生物碱:青藤碱(1),sinomenine N-oxide(2)和青风藤碱(3);4个原小檗碱型生物碱:四氢巴马汀(4),紫堇单酚碱(5),千金藤定碱(6)和异紫堇单酚碱(7)。其中化合物2和7为首次从地不容中分离得到,化合物3对乙酰胆碱酯酶具有明显的抑制作用(IC50=3.03μM)。
