高效液相色谱法对赤砂糖水解液中三种物质的分离和测定

         

摘要

建立了一种同时分离、测定赤砂糖水解产物中的乙酰丙酸(LA)、5-羟甲基糠醛(5-HMF)和甲酸(FA)的高效液相色谱法.采用Agilent TC-C18色谱柱(4.6×250mm,5μm)、Agilent 1200 series VWD紫外检测器,以乙腈:磷酸-磷酸二氢钠缓冲溶液(pH2.6)=15∶85(体积比)为流动相进行分离,检测波长210nm,柱温30℃,流速1 mL/min.在此条件下,三种物质的色谱峰面积与浓度线性良好,线性相关系数r在0.9993~0.9999之间,平均回收率为99.02%~106.28%,精密度实验的相对标准偏差为0.25% -1.30% (n=5).%A high performance liguid chromatographic( HPLC) method was established for the separation and determination of levu-linic acid( LA) ,5-hydroxyinethyl furfural( 5-HMF) and formic acid( FA) in the hydrolysate of brown sugar. The hydrolysate of brown sugar was separated on an Agilent TC-C18(4.6x250mm,5μm)coluinn by the buffer solution of acetonitrile:phosphoric acid-sodium dihydrogen phosphate(pH2.6)= 15:S5(v/v)as mobile phase at a flow rate of lmL/min. The samples were detected by Agilent 1200 series VWD UV detector with the wavelength of 210nm at 30%. Under the conditions,good linear relationships existed be+ tween peak area and concentration of three components in the standard mixed solution, the linear correlation coefficients were 0.9993-0.9999, the average recoveries were 99.02%-106.28% , and the relative standard deviation of the precision test were 0.25%-1.30%(n=5).
