唾液乳杆菌降解4-硝基喹啉-1-氧化物所需条件的研究

         

摘要

4-Nitroquinoline-1-Oxide (4-NQO) is one kind of genotoxins. Lactobacillus salivarius FDB89, screened from Guangxi Bama longevity feces, could degrade 4-NQO to nontoxic substance. Different concentrations of L. salivarius FDB89 were added in reaction solution with 4-NQO (20mg/L). Samples were detected by high performance liquid chromatography (HPLC). The results showed that L. salivarius FDB89 could not degrade 4-NQO when its OD600 was lower than 2.3(about 2.1×1011cfu/ml). When the concentration was about 1.4×1013cfu/ml, the degradation reaction was obvious and several intermediate metabolites or degradation products were detected. The degradation ability of L. salivarius FDB89 against 4-NQO had positive correlation with its density.%4-硝基喹啉-1-氧化物(4-NQO)是一种基因毒素.广西巴马老人源唾液乳杆菌FDB89可降解4-NQO为无毒物质.将不同浓度的唾液乳杆菌FDB89菌悬液与4-NQO溶液(4-NQO终浓度为20mg/L)混合,反应一定时间后分别取样,用高效液相色谱法(HPLC)检测其中的4-NQO含量变化情况.唾液乳杆菌FBD89菌体浓度达到OD600=2.3(约2.1×1011cfu/mL)时,降解现象比较明显,并在菌体浓度约为1.4× 1013cfu/mL时能检测到4-NQO的降解产物.研究结果表明,唾液乳杆菌FBD89具有降解4-NQO的能力,降解能力随菌体浓度升高而提高.
