鱼油对老年Ⅰ期压疮模型大鼠免疫功能、炎症反应及细胞凋亡的影响

         

摘要

目的 探讨鱼油对老年Ⅰ期压疮大鼠细胞免疫、炎症反应及细胞凋亡的影响.方法 随机将20只老年SD大鼠分成三组,正常组和模型组各6只,采取普通饲料喂养,鱼油干预组8只给予普通饲料加1.6 g/(kg·d)鱼油灌胃喂养20 d.正常组不受压,模型组和鱼油干预组承受22.47kPa压力2h后三组大鼠一起实施安乐死.采集受压组织中心标本,行常规石蜡切片、HE染色及TUNEL荧光标记;光镜下观察三组大鼠皮肤肌肉组织病理变化;检测10%匀浆肌组织中白细胞介素-2(IL-2)、白细胞介素-6 (IL-6)、一氧化氮(NO)、一氧化氮合酶(NOS)的含量和活性及细胞凋亡指数(AI)的变化.结果 受压组织病理形态学显示,与模型组相比,鱼油干预组受压表皮及组织炎症细胞浸润少,受压组织受损程度明显减轻(P=0.00).与正常组比较,模型组IL-2、IL-6、NO、NOS及AI水平均显著增高(P=0.00);与模型组比较,鱼油干预组IL-2、IL-6、NO、NOS及AI水平均显著降低(P=0.00).结论 鱼油的早期应用可有效抑制受压组织的免疫、炎症反应和细胞凋亡,从而降低老年压疮发生率.%Objective To investigate the impact of fish oil on the immunity,inflammation and apoptosis in stage Ⅰ elderly pressure sores model rats.Methods 20 adult SD rates were randomly divided into three groups:the normal group with 6 rates,the model group with 6 rates and the fish oil intervention group with 8 rates.The normal group and the model group were given common fodder and the fish oil intervention group was given common fodder plus gavage of 1.6g/(kg·d) fish oil for 20 days.The normal group was not given pressure,but the model group and the fish oil intervention group were given 22.47 kPa pressure for 2 h,after which all the 3 groups of rats were euthanized together.The pressurized tissue center specimens were collected for conventional paraffin section,HE staining and TUNEL fluorescent marker.The pathological changes of the rats' skin and muscle tissues were observed under light microscope.The changes of contents and activity of interleukin-2 (IL-2),interleukin-6 (IL-6),nitric oxide (NO) and nitric oxide synthase (NOS) and the apoptosis index (AI) in the 10% homogenate muscle tissue were detected.Results The pathomorphological results showed that compared to the model group,the fish oil intervention group had reduced inflammatory cellular infiltration in the pressurized surface skin and tissue,and the damage degree of pressurized tissue significantly reduced (P=0.00).Compared to the normal group,the model group had significantly increased IL-2,IL-6,NO,NOS and AI levels (P=0.00); compared to the model group,the fish oil intervention group had significantly reduced IL-2,IL-6,NO,NOS and AI levels (P=0.00).Conclusion Early application of fish oil can effectively inhibit the immune,inflammatory reaction and apoptosis of pressurized tissue,thereby reducing the incidence of senile pressure sores.
