大鼠脑挫伤后细胞间黏附因子1时序性表达的实验性研究

         

摘要

Objective To investigate the changes of intercellular adhesion molecule-1 expression in the different posttrau-matic intervals after cerebral contusion in rats. Methods Based on Feeney's method, the rat model of cerebral contusion was established. The samples taken at 1 h, 24 h, 48 h, 72 h, 7 d after cerebral contusion and stained by western blot, the eight normal rats was choose as controls. Results The expression of ICAM-1 protein was observed at th after injury, increased at 24 h, reached the peak at 72h and then was decreased. The expression of ICAM-1 in the experimental group on the 7th day after injury still was significantly higher than that in the control group. Each experimental group compared to control group, the differences had statistical significance (P < 0.05). Significant statistic difference was found in multiple comparison (P < 0.05). Conclusion The sequential expression of ICAM-1 is used for the estimation of early posttraumatic intervals after cerebral contusion in forensic practice.%目的 观察大鼠脑挫伤后不同时间段细胞间黏附因子1 (intercellular adhesion molecule-1,ICAM-1)的表达变化规律.方法 参照Feeney法建立大鼠闭合性脑挫伤模型,将实验大鼠分为对照组和脑挫伤后1h、6h、24h、48 h、72 h、7d组,共7组,每组8只,运用Western blot方法检测脑挫伤组织中ICAM-1的表达.结果 大鼠脑挫伤1h可见ICAM-1有少量表达,伤后24h表达明显增多,72 h达到高峰,其后下降,伤后7d仍有表达,且高于对照组水平.各实验组与对照组比较,差异均有统计学意义(均P< 0.05),各实验组两两比较差异均有统计学意义(均P<0.05).结论 大鼠脑挫伤后ICAM-1表达随着时间的变化表达呈现一定的规律性,可为推断脑挫伤早期损伤时间提供一定的参考依据.
