羟基喜树碱对人乳腺癌MCF-7细胞增殖及凋亡的影响

         

摘要

Objective: To explore the effect of Hydroxycamptothecin (HCPT) on the proliferation and apoptosis of human breast cancer cell line MCF-7 in vitro, and to provide experimental basis for the clinical medication of breast cancer.Methods: HCPT with different concentrations (10 μmol/L, 50 μmol/L, 100 μmol/L, 200 μmol/L) were used to treat MCF-7 cells for 48 hours, and MCF-7 cells without HCPT were served as the control group. MTT assay was applied to determine the proliferation of MCF-7 cells, Hoechst33258 staining was used to detect the apoptosis of MCF-7 cells, and RT-PCR was used to detect the effect of HCPT on the expression of Casepase-3 mRNA in MCF-7 cells. Results: Compared with the control group, 50 μmol/L 100 μmol/L and 200 μmol/L HCPT showed obvious inhibitory effect on MCF-7 cells (P<0.05, P<0.01), and the inhibitory effect of 200 μmol/L was the most obvious, the inhibitory rate showed a concentration-dependent tendency. Fluorescence microscope observation showed that typical apoptosis morphological changes could be observed in 100 μmol/L and 200 μmol/L HCPT treated cells. Compared with the control group, the expressions of Caspase-3 mRNA significantly increased in HCPT treated cells (P<0.05. P<0.01), and it also showed a concentration-dcpendent tendency. Conclusion: HCPT can inhibit the proliferation of MCF-7 cells in vitro, and induce cell apoptosis by Caspase-3 pathway, and further achieve its role of inhibiting the growth of tumor cells.%目的:探讨羟基喜树碱在体外对人乳腺癌MCF-7细胞增殖及凋亡的影响,为乳腺癌的临床用药提供实验依据.方法:将不同浓度(10μmol/L、50μmol/L、100μmol/L、200μmol/L)的羟基喜树碱作用于人乳腺癌MCF-7细胞48 h,并设不加羟基喜树碱溶液的MCF-7细胞作为对照组,用MTT法测定MCF-7细胞的增殖情况,用Hoechst33258染色检测MCF-7细胞的凋亡情况.以RT-PCR检测羟基喜树碱对MCF-7细胞Casepase-3 mRNA表达的影响.结果:与对照组比较,50 μmol/L、100μmol/L及200μmol/L羟基喜树碱对细胞增殖均有抑制作用(P<0.05、P<0.01),尤以200μmol/L的抑制作用最明显,其抑制率呈浓度依赖性增加.荧光显微镜观察显示,100μmol/L及200μmol/L羟基喜树碱处理组中可见典型的凋亡形态学改变.与对照组比较,各羟基喜树碱处理组中MCF-7细胞Caspase-3 mRNA表达水平均升高(P<0.05、P<0.01),并呈浓度依赖性升高趋势.结论:羟基喜树碱可在体外抑制MCF-7细胞增殖,并通过Caspase-3途径诱导细胞凋亡,达到抑制肿瘤细胞生长的作用.
