目的:探索快速、经济的银染色方法.方法:本研究对前人文献所报道的一种银染检测方法的检测步骤和试剂用量进行了优化,省略了操作过程中的固定步骤,简化了停显液的组成成分.结果:优化后的方法在配制试剂时只需3种化学试剂,染色液中硝酸银的用量被减少至1.0 g/L,显影液中氢氧化钠的浓度被降低至10 g/L,DNA检测效果表明优化前后的两种检测方法的检测灵敏度均可达5 ng左右.结论:优化后的DNA银染色检测方法更加经济、快速.%Objective: To establish a cheaper and faster silver staining method. Methods: One silver staining method reported by other researchers had been modified in the steps and reagent dosages, in which, the fixaLion step had been omitted, the composition of stop solution was simplified. Results: Only three kinds of chemical reagents were required for detecting solutions, the dosage of sivler nitrale in staining solution and sodium hydroxide in developing solution were reduced to 1.0 g/L and 10 g/L respectively. The limit of DNA detection upon visual observation for both of the two methods was approximately 5 ng. Conclusion: The optimized silver staining method is much cheaper and faster.
