目的:建立测定三花膏中蒙花苷含量的高效液相色谱(HPLC)法。方法以三花膏中野菊花药材中的主要活性成分蒙花苷为对照品,采用Phenomenex Luna C18反相色谱柱(250 mm ×4.6 mm,5μm ),以甲醇( A )-含10 mmol/L H3PO4的水溶液( B,48:52)为流动相进行等度洗脱,流速为1.0 mL/min,检测波长为334 nm,柱温为35℃。结果蒙花苷质量浓度在4.67~46.74 g/L范围内与峰面积线性关系良好( r=0.9999,n=5),加样回收率为99.75%,RSD为1.80%( n=6)。结论该方法简便、准确,专属性强,适用于三花膏中蒙花苷的含量测定。%Objective To develop an HPLC method for the content determination of linarin in Sanhua Ointments. Methods Linarin, which was the main active ingredient of Flos Chrysanthemi Indici in Sanhua Ointments, was used as the reference standard. The isolation was performed on a Phenomenex Luna C18 reversed phase column ( 250 mm × 4. 6 mm, 5 μm ) with the mobile phase consisting of methanol-water ( containing 10 mmol/L H3PO4 ) ( 48 :52 ) , isocratic elution;the flow rate was 1. 0 mL/min, the detection wavelength was 334 nm and the column temperature was set at 35 ℃. Results The linearity of linarin was good in the range of 4. 67-46. 74 g/L ( r=0. 999 9, n=5 ) , the average recovery rate was 99. 75% and RSD was 1. 80%( n=6). Conclusion This method is simple, accurate, and specific for the content determination of linarin in Sanhua Ointments.
