洛铂对肺癌A549细胞作用的初步探讨

         

摘要

Objective To explore the effect of lobaplatin on lung cancer cell line .A549. Methods A549 cells in log phase were collected and derided into negative control group and experimental groups. Experimental groups were treated with different concentrations of lobaplatin(2.5, 5, 10, 20 and 40 μmol/L). MTI colorimetry and flow cytometry were used to detect the effect of lobaplatin on lung cancer cell proliferation inhibition, cell cycle arrest, induction of apoptosis and so on. The application of Western blotting was used to detect the expression of Bcl-2 in A549 cell which was treated by lobaplatin. Results Lobaplatin could inhibit the proliferation and apeptosis of lung cancer cell line A549. The inhibition was in concentration and time dependent manner. Sub-G1 apoptotic peak appeared after lobaplatin treatment for 24h. Western blotting results showed that Bcl-2 protein level decreased after lobaplatin treatment. Conclusion Lobaplatin can significanfiy inhibite the proliferation of A549 lung cancer cell and induce apoptosis and the change of cell cycle primarily through the regulation of Bcl-2 protein expression.%目的 探讨洛铂对肺癌A549细胞增殖与凋亡的作用及其机制.方法 取对数生长期的肺癌A549细胞进行实验,分为阴性对照组和实验组(加入不同浓度洛铂:2.5、5、10、20和40μmol/L).采用MTT比色法、流式细胞技术检测洛铂对肺癌细胞的增殖抑制、细胞周期阻滞、细胞凋亡诱导等作用,应用蛋白印迹法分析洛铂对A549细胞Bcl-2蛋白表达的影响.结果 洛铂能够抑制肺癌A549 细胞的增殖并诱导其凋亡,并呈浓度和时间依赖;流式细胞检测示细胞周期G1期前有低于二倍体细胞的凋亡峰;蛋白印迹检测结果显示洛铂可使A549细胞Bcl-2蛋白表达下降.结论 洛铂能显著抑制肺癌A549 细胞增殖,诱导细胞凋亡和细胞周期变化,可能与调节A549细胞Bcl-2蛋白的表达有关.
