金匮肾气丸拮抗环磷酰胺对大鼠生殖毒性的实验研究

         

摘要

目的 探讨金匮肾气丸对腹腔注射环磷酞胺(CTX)雄性SD大鼠生殖功能的影响.方法 取15月龄雄性SD大鼠40只,随机分成模型组和金匮肾气丸高、中、低剂量治疗组,每组均10只,每组大鼠均给予腹腔注射环磷酰胺混悬液(1ml/kg·d-1,qd)连续5d.模型组以蒸馏水灌胃,治疗组以金匮肾气丸混悬液高、中、低剂量灌胃,连续28d.末次给药24h后处死动物,测睾丸、附睾的湿重:观察精子密度、活力和活率.结果 用药后,金匮肾气丸组大鼠睾丸及附睾脏器系数与模型组相比显著提高(P<0.01):与模型组比较,金匮肾气丸各组精子密度明显升高(P<0.05);与模型组比较,高剂量组精子活力升高(P<0.05),而中、低剂量组则差异无统计学意义,P>0.05;与模型组比较,中、高剂量组精子活率明显升高(P<0.05),其中,以高剂量组最为明显(P<0.01),而低剂量组则差异无统计学意义.结论 金匮肾气丸可以提高环磷酞胺染毒大鼠的睾丸、附睾脏器系数,提高精子密度、活力和活率.金匮肾气丸可以有效拮抗环磷酞胺对SD大鼠的生殖毒性,保护其生殖功能.%Objective To investigate the effects of Jinkuishenqi Pill on reproductive function of rats injected by cyclophosphamide (CTX). Methods Forty 15-month-old male SD rats were randomly divided into the model group, the high-dose treatment group, the medium-dose treatment group and the low-dose treatment group. There was 10 rats in each group. All rats were treated with intraperitoneal injection of cyclophosphamide suspension (1 ml/kg · d-1, qd) for 5 days. Rats in the model group were orally administrated with distilled water. Rats in different treatment groups were respectively administrated with low-, medium- and high-dose Jinkuishenqi Pill suspension for 28 days. All rats were sacrificed after 24-hour at the last dose Wet weight of the testis and epididymis were measured and the sperm density, vitality and viability were detected. Results After administration, the organ coefficient of testis and epididymis of rats in the treatment groups was improved significantly (P<0.01). Compared with that of the model group, the sperm density of the treatment groups were significantly increased (P<0.05). Sperm motility increased most significantly in the high-dose treatment group (P<0.05), sperm viability increased most significantly in medium-dose treatment group (P<0.05) and the high-dose treatment group (P<0.01), compared with that in the control group. Conclusion Jinkuishenqi Pill could improve the organ coefficient of testis and epididymis of rats injected by cyclophosphamide, and promote sperm density, vitality and viability. Jinkuishenqi Pill can effectively antagonize the reproductive toxicity caused by cyclophosphamide and protect reproductive functions.
