益气养阴通络方对糖尿病足小鼠VEGF、胰腺氧化应激、胰岛形态的影响

         

摘要

目的:观察益气养阴通络方对糖尿病足(diabetic foot,DF)小鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)及胰腺抗氧化应激能力、胰岛形态的影响.方法:SPF级C57 BL/6J雄性小鼠70只,随机抽10只为正常组,其余小鼠腹腔注射链脲佐菌素(Streptozotocin,STZ) (120mg/kg)及结扎股动脉、股静脉建立DF模型,分为模型组、二甲双胍+西洛他唑组(西药对照组)、通塞脉片组(中药对照组)、益气养阴通络方高、中、低剂量组,连续用药3周,检测干预前后各组小鼠体质量、空腹血糖(fasting blood glucose,FBG)、干预后血清VEGF、胰腺组织超氧化物歧化酶(Superoxide dismutase,SOD)、还原型谷胱甘肽(Glutathione,GSH)活性及丙二醛(Malondialdehyde,MDA)含量.实验末摘取胰腺,苏木精-伊红(hematoxylin-eosin,HE)染色观察胰岛结构.结果:益气养阴通络方能显著降低FBG,上调VEGF,提高胰腺SOD、GSH活性,降低MDA含量.胰岛HE染色提示益气养阴通络方高剂量组形态学改变优于模型组.结论:益气养阴通络方能降低血糖,调节血清VEGF浓度,增强胰腺抗氧化应激能力,改善胰岛病理形态,从而促进血管再生,阻止或延缓胰腺功能的损害,加速足部溃疡愈合.%Objective:To observe the effect of Yiqi Yangyin Tongluo Formula on vascular endothelial growth factor (VEGF),pancreatic antioxidative ability and islet morphology of diabetic foot (DF)mice.Methods:10 mice were randomly selected from 70 SPF male C57BL/6J mice.Other mice,given 120mg/kg streptozotocin (STZ) injection and femoral artery and vein ligature to establish DF models,were divided into model group,Metformin + Clostazol group,Tongsaimai group,and high,middle and low dose of Yiqi Yangyin Tongluo Formula groups.During gavage for 3 weeks,body weight and fasting blood glucose (FBG) of all the mice were observed dynamically,and VEGF,superoxide dismutase (SOD),glutathione(GSH) and malondialdehyde (MDA) were determined after gavage.At the end of the experiment,pancreases were removed and Hematoxylin-eosin (HE) staining was used to observe the structure of islets.Results:Yiqi Yangyin Tongluo Formula can lower FBG,raise VEGF level,SOD and GSH activity,and decrease MDA level.HE staining showed that the morphological changes of the high dose of Yiqi Yangyin Tongluo Formula group were better than those of model group.Conclusion:Yiqi Yangyin Tongluo Formula can lower glucose,regulate VEGF level,enhance pancreatic antioxidative ability and improve islet morphology to promote revascularization,prevent or delay pancreatic function lesion and thus accelerate foot ulcer healing.
