杜仲叶提取物对体外培养的成骨细胞代谢功能调节研究

         

摘要

Objective:Throuth studying three abstractive parts from Duzhong leaves with different concentration affecting osteoblast cultured in vitro,We can make it cleaver the action principle of Duzhong in tonifying kidney,invigorating channeis and collaterals,and reinforcing bone,as well as screening out the effective parts of Duzhong leaves in pleventing and treating osteoporosis.Method:Three parts(Ⅰ、Ⅱ、Ⅲ)abstracting from Duzhong leaves with different concentration affect osteoblast cultured in vitro,skull-cap bone of new born SD rat was used for primary culturre,and the secondary culture cell were used in experiment. Determented indexes: (1) cell proliferation:MTT calcalating method. (2) vigor of Alkaline phosphatase(ALP):p-nitroghosphate method. Result:Cell proliferation in part Ⅱ & Ⅲ groups were increased significantly (P<0.001),compared with control group. Vigor of ALP were increased in all three parts (P<0.001) comparaed with control groulp. Conclude:In regalating bone metabolism effect of part Ⅱ & Ⅲ with 10-5mg/L high concentration are most obvious.%目的:研究杜仲叶提取物对体外培养成骨细胞(Osteoblast OB)代谢功能的调节作用,从而进一步明确杜仲补肾、强筋健骨的作用机理以及筛选杜仲叶防治骨质疏松药效成分。方法:用杜仲叶三个提取部位(Ⅰ、Ⅱ、Ⅲ)不同浓度对体外培养的成骨细胞作用。由新生SD大鼠头盖骨作原代培养,用传代的第一代细胞做药效试验。测定指标:(1)细胞增殖情况,用MTT计数法[1、2];(2)碱性磷酸酶(ALP)活性测定,对硝基磷酸盐法。结果:Ⅱ、Ⅲ提取部位对细胞增殖作用与对照组比较P<0.001,ALP的活性与对照组比较,三个部位均为P<0.001。结论:表明Ⅱ、Ⅲ提取部位的10-5mg/L浓度组具有明显的调节骨代谢功能的药效作用。
