乳腺癌中微管相关蛋白LC3A和LC3B的表达及意义

         

摘要

Purpose To investigate the expression levels of isoforms of microtubule-associated protein 1 light chain 3 ( LC3A and LC3B ) and their mRNA in breast carcinoma,and to explore the relationship hetween LC3A and LC3B and the pathogenesis and metastasis of breast carcinoma. Methods SP immunohistochemical and RT-PCR were used to detect the expression levels of LC3A, LC3B and their mRNA in the normal breast tissues( 8 cases ) , breast proliferative diseases( epithelial hyperplasia )( 20 cases ),breast preinvasive carcmoma( 10 cases )and breast invasive carcinoma( 40 cases ). Results ( 1 )There was significant relationship between LC3A and LC3B staining ( intensity and proportion ) and type of tissues;( 2 )Both LC3A and LC3B mRNA were showed significant increasing gradient of expression levels in the normal breast tissues, breast proliferative diseases( epithelial hyperplasia ) ,breast preinvasive carcinoma and breast invasive carcinoma. Based on WHO grade, LC3A mRNA levels were showed gradual increase in cancer specimens with grades Ⅰ , Ⅱ and Ⅲ ( P < 0. 05 ); ( 3 )There was no significant relationship between LC3 expression and clinical stage, age group and lymph node metastasis. Conclusions LC3A protein expression presents in greater ahundance and in a higher percentage of cells in breast cancer tissues as compared with the normal tissue, and LC3A mRNA levels show gradual increase in cancer specimens with grades Ⅰ , Ⅱ and Ⅲ , suggesting that LC3A isform may be related to the occurrence and development of human breast cancer.%目的 探讨微管相关蛋白MAP1LC3(LC3A和LC3B)在乳腺癌组织中的表达,阐明其与乳腺癌发生、发展及转移的关系.方法 以8例正常乳腺组织为对照,采用免疫组化SP法和半定量RT-PCR技术检测在乳腺增生性病变(20例),乳腺非浸润性癌(10例)、乳腺浸润性癌(40例)中MAP1LC3A、MAP1LC3B及其mRNA的表达水平.结果 (1)LC3A和LC3B着色强度及着色细胞百分比和不同组织类型有重要关系.(2)LC3A与LC3B mRNA在正常乳腺组织、乳腺增生性病变、乳腺非浸润性癌和乳腺浸润性癌4组病例中的表达依次呈递增趋势,差异具有统计学意义(P<0.05).根据不同的组织学分级,LC3A在Ⅰ级、Ⅱ级、Ⅲ级中的表达呈递增趋势,且差异具有统计学意义(P<0.05).(3)LC3A、LC3B表达与临床分期、患者年龄、淋巴结转移之间差异均无显著性.结论 与正常组织相比,LC3A蛋白在乳腺癌组织中表达更多的强阳性和更高的阳性细胞百分比,并且在WHO分级中从Ⅰ~Ⅲ级,其mRNA表达依次呈递增趋势,由此提示LC3A亚型可能与乳腺癌的发生发展有关.
