血必净对脂多糖诱导急性肺损伤大鼠的保护作用

         

摘要

Objective To explore the potential protective effect of Xuebijing on lipopolysaccharide-induced acute lung injury(ALI) in rats.Methods 45 Wistar rats were randomly divided into 3 groups as following:(1)con-trol group(n=15),(2)LPS group(n=15),(3)Xuebijing treatment group(XBJ group) (n=15).LPS(5mg/kg) was infused by femoral vein to establish ALI model in LPS group and XBJ group .The rats were received XBJ at a dose of 4ml/kg ( XBJ group) and the equivalent volume normal saline (control/LPS group) 30 min before the intravenous injection of a bolus of LPS .Five rats in each group were sacrificed on the 6th、12th and 24th day after the model , which were divided into subgroup I ,Ⅱ,and Ⅲ.The pathomorphism changes of lung tissue were observed using light microscope ,and the dynamic arterial blood gas analysis ,were conducted .The lung wet to dry weight ratio ( W/D) and the semi-quantitative lung pathologic scores was also calculated .The expression of TNF-αwere determined by enzyme-linked immunosorbent assay(ELISA).Results In comparison with the LPS group,the arterial partial pressure of oxy-gen(PaO2) was significantly increased(P<0.05),and the ratio of lung tissue wet /dry (W/D),the scores were sig-nificantly decreased(P<0.05) in XBJ group on 6 th,12 th,24th day after LPS injection.The levels of serum TNF-a were significantly decreased(P<0.05).Conclsion The treatment with Xuebijing could alleviate the inflammatory reaction and protect against the septic lung injury induced by LPS ,which maybe involved in the down-regulation of TNF-α.%目的:观察血必净对脂多糖( LPS)诱导的急性肺损伤( ALI)大鼠的保护作用。方法 Wistar大鼠随机分为三组:对照(NS)组(15只)、脂多糖(LPS)组(15只)、血必净(XBJ)组(15只),LPS组和XBJ组静脉注射LPS(5 mg/kg)建立大鼠ALI模型;XBJ组在造模前半小时静脉注射血必净4 mL/kg,NS组和LPS组则给予等容积0.9%氯化钠注射溶液;在造模后6 h、12 h、24 h各组分别随机处死大鼠5只,分别作为I、Ⅱ、Ⅲ亚组,观察肺组织形态学变化,测定各组大鼠不同时间点动脉血气分析、肺组织湿/干重比( W/D)和血清肿瘤坏死因子-α( TNF-α)表达。结果与LPS组相比,在LPS注射后6 h、12 h和24 h各时间点,XBJ组血氧分压(PaO2)较高(P <0.05),而肺组织W/D比值、肺组织病理评分、血清TNF-α水平显著降低(P <0.05)。结论血必净预处理可以使LPS诱导ALI大鼠的肺组织水肿和炎症细胞浸润的程度减轻,可能与降低TNF-α有关。
