胰高血糖素样肽-1受体在糖尿病大鼠周围神经中的表达及格列齐特的干预作用

         

摘要

目的 探讨胰高血糖素样肽-1受体(GLP-1R)在糖尿病大鼠周围神经中表达的意义及格列齐特的干预作用.方法 采用高脂饲料和小剂量链脲佐菌素( STZ)注射相结合的方法,制备2型糖尿病(DM)大鼠模型.应用HE染色在光镜下观察各组大鼠坐骨神经形态学动态变化,应用免疫组织化学方法检测各组GLP-1R、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的蛋白表达.结果 HE染色显示DM大鼠在前4周,坐骨神经即出现形态学改变,且损伤程度随病程的延长而加重.格列齐特治疗8周组较同期DM组损伤明显减轻.DM组与正常组(NC)相比,GLP-1R蛋白表达明显下降,Caspase-3蛋白表达明显升高.格列齐特用药组与DM组相比,GLP-1R蛋白表达明显升高,Caspase-3蛋白表达明显下降.结论 GLP-1R的表达变化与DM大鼠的坐骨神经病变程度有关;格列齐特可提高GLP-1R表达,减少Caspase-3表达.%Objective To investigate the expression and effect of glucagon-like peptide-1 receptor (GLP-1R) in sciatic nerve in diabetic rats and the intervention affect of gliclazide. Methods The rat model of type 2 diabetes was made by feeding a diet enriched with energy and ip low-dose streptozotocin. Histopathological changes of sciatic nerve in each group were observed by light microscope with HE dye. Expressions of GLP-1 R and caspase-3 in each group were detected by the immunohistochemistry. Results In DM rats,there was morphological changes of sci-atic nerve in the early 4 weeks by HE dye method, and the damage aggravated with the extension of DM course. The changes in gliclazide treated group were slighter than those of DM 8 weeks group in the corresponding period. In DM group,the expression of protein CLP-1R decreased significantly and the expression of protein Caspase-3 increased significantly in comparison with the NC group. In the treated groups,the expression of protein GLP-1R increased significantly and the expression of protein Caspase-3 decreased significantly in comparison with the DM group. Conclusion The changes of GLP-IR expression is related to the lesion degree of peripheral nerve in diabetes, gliclazide can increase GLR-K expression and inhabit the Caspase-3 expression.
