目的 观察大鼠心肌梗死后血浆中miR-214的表达变化.方法 10只雄性SD大鼠,通过前降支动脉结扎法建立大鼠急性心肌梗死模型,模型制备成功后随机分为心肌梗死后14 d组、28 d组,另设对照组,每组5只动物.抽提血浆中的miRNA,应用荧光定量PCR法检测miR-214的表达.结果 与假手术组相比,术后14 d组miR-214的表达增加60%[(1.60±0.09)vs.(1.00±0.06),P＜0.01],术后28 d组miR-214的表达增加116%[(2.16±0.13)vs.(1.00±0.06),P＜0.01].结论 大鼠心肌梗死后随着心肌梗死时间的延长,血浆中miR-214的表达逐渐上调,miR-214可能参与调控了心肌梗死后心室重塑.%Objective To detect the change of expression level of plasma miR-214 in rats with post acute myocardium infarction. Methods Rats underwent left descending coronary ligation or sham surgery. Rats with MI was assigned to two groups ( n = 5 ). Real-time PCR was developed to detect the expression of miR-214 in plasma. Results The expression level of miR-214 in plasma was up-regulated in the 14th and 28th day post MI. Compared to sham group, the expression level of miR-214 increased by 60％ ( 1.60 ± 0.09 vs 1.00 ± 0.06,P ＜ 0.01 ) in the 14th day and by 116％ (2.16 ±0.13 vs 1.00 ±0.06,P ＜0.01) in the 28th day post MI. Conclusion The expression of plasma miR-214 increases with ventricular remodeling post MI in rat. MiR-214 may be involved in ventricular remodeling post MI.
