胃电起搏对大鼠胃窦Cajal间质细胞的影响

         

摘要

目的 研究胃电起搏对大鼠Cajal间质细胞(ICC)数量和C-kit受体表达的影响,探讨ICC在胃电起搏调控胃慢波活动中的作用.方法 建立Wistar大鼠胃起搏模型,将大鼠分为起搏组(n=8)和对照组(n=8).选用适宜的起搏参数以控制胃电慢波,持续刺激1 h,连续刺激20 d后处死大鼠并取胃窦组织,以免疫组织化学方法结合图像分析技术和Western blot方法分析C-kit受体免疫反应阳性细胞和C-kit蛋白表达变化.结果 起搏组大鼠胃肌间神经丛内C-kit免疫反应阳性产物面积和平均光密度值均显著高于对照组[(1976.76±336.67)vs (1368.23±459.32),P<0.05;(0.19±0.50)vs (0.12±0.52),P<0.05],刺激组大鼠胃窦组织C-kit蛋白表达显著增高[(0.50±0.09)vs(0.29±0.02),P<0.05].结论 胃电起搏后胃窦肌间神经丛ICC数量增多,参与了胃电起搏调控胃慢波活动.%Objective To investigate the effect of gastric electrical pacing on interstitial cells of Cajal (ICC)in the antral myenteric plexus in rats. Methods The Wistar rat models with two pairs of electrodes were established by operation, and were divided into two groups, i. e. pacing group ( GES, n = 8 ) and control group ( n = 6 ). Pacing group was given long pulse GES for 1 hour daily for 20d, while the control was allowed free access to food and water.On the 20th day,the rats in both two groups were sacrificed. The number of ICC in the antral myenteric plexus was counted after immunohistochemical staining for c-Kit. C-kit protein expression was studied by western blot analysis. β-actin,a steadily expressed protein,was used as the internal reference, and the C-kit protein to β-actin protein in the integrated optical density was calculated to represent the relative expression amount of C-kit protein. Results C-kit immunoreactivity in the antral myenteric plexus in GES group was more abundant than that of control ( 806.421 ±342. 135 vs 448.3 ±261. 467 ,P <0.05). The expressions of C-kit protein were increased in GES group compared with that of control (0.50 ± 0.09 vs 0.29 ± 0.02,P < 0.05 ). Conclusion ICC in the antral myenteric plexus may play an important role in the gastric electrical pacing.
