Objective:To investigate the changes of aquaporin-5(AQP5) and mucin 5AC(MUC5AC) in SPC-A1 cell line stimu-lated by lipopolysaccharide (LPS). Methods: After SPC-A1 cell line was stimulated by LPS,the mRNA levels of AQP5 and MUC5AC were detected by real-time polymerase chain reaction (PCR),and AQP5 and MUC5AC protein were measured by Western blot and enzyme-linked immunosorbent assay (ELISA). Results; The protein expression and transcription of AQP5 decreased while the MUC5AC expression increased in a dose (0 μg/mL、5μg/mL、10 μg/mL,20μg/mL and 40μg/mL,respectively) and time-dependent manner by LPS stimulation in SPC-Al cell line. Conclusions: Decreased AQP5 expression was correlated with increase of MUC5AC in SPC-Al cell line after LPS exposure.%目的:探索脂多糖( lipopolysaccharide,LPS)刺激下水通道蛋白5(aquaporin-5,AQP5)和黏蛋白5AC(mucin 5AC,MUC5AC)的变化.方法:采用不同的时间点、不同浓度LPS干预SPC-A1细胞株,荧光定量聚合酶链反应(polymerase chain reaction,PCR)检测AQP5和MUC5AC的mRNA水平,Western杂交、酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法检测AQP5和MUC5AC蛋白的变化.结果:给予SPC A1细胞LPS 0μg/mL、5 μg/mL、10 μg/mL、20μg/mL、40 μg/mL浓度刺激后,AQP5mRNA和AQP5蛋白表达呈LPS浓度依赖性降低,而MUC5AC mRNA和MUC5AC蛋白表达呈LPS浓度依赖性升高.LPS干预0、12、24、48 h后,AQP5 mRNA和AQP5蛋白呈时间依赖性降低,MUC5AC mRNA和MUC5 AC蛋白升高,两者呈负相关.结论:LPS刺激后AQP5和MUC5AC的变化呈负相关.
