用组织工程的方法建立人牙髓细胞体外三维立体培养模型

         

摘要

AIM: Three-dimensional cultured model of pulp cell was established to provide a tool in the research of pulp disease. METHODS: Pulp cells were cultivated in vitro. Cells were Collected and seeded on three-dimensional framework of polyglycolic acid (PGA). Inverted microscope, scanning electronic microscope and transmission microscope were used to observe the cell growth in the scaffold. RESULTS: Cells adhered on the fiber of PGA after 24 hours, then began to proliferate and grow in the framework of three-dimension. Scanning electronic microscopy showed that cells with spindle shape spread on PGA fiber. Transmission microscopy showed that the microstructure of cell was normal. CONCLUSION: Cells seeded on PGA framework was an efficient way to establish three-dimensional cultured model of pulp cells.%目的:建立人牙髓成纤维细胞体外三维立体培养模型,为研究牙髓生理病理奠定基础。方法:组织块法培养人牙髓细胞,扩增后,接种于聚乙醇酸(PGA)三维支架上,继续培养,通过倒置显微镜、扫描电镜和透射电镜观察细胞的生长情况。结果:细胞接种24h后开始贴附于PGA纤维上,然后开始伸展并增殖,PGA纤维上的细胞不断增长。扫描电镜可见细胞在PGA纤维上生长良好,呈长梭形。透射电镜观察表明PGA上的细胞超微结构正常。结论:将牙髓细胞接种于PGA无纺网上,可以有效地建立牙髓细胞三维立体培养模型。
