摘要

Objective To study the expression and mechanism of several cytokines in the fracture of the condyle.Methods fibroblast growth factor 1 (FGF1),insulin-like growth factor 1 (IGF1),interleukin (IL)-1β,IL-6,transforming growth factor (TGF)-β and bone morphogenetic protein (BMP) were determined by immunohistochemical method.Phosphatidylinositol 3 kinase (PI3K),protein kinase B (Akt) protein and mRNA expression were detected in two groups.Results BMP:Mean standard deviation of the expression side was (101.51 ± 2.16) cm2,the control side was (101.23 ±1.90) cm2,the expression of BMP was not significantly different from the control side and the experimental side (P > 0.05).There was no significant difference in the expression of FGF1 (P > 0.05) between the control side[(91.03 ± 2.37) cm2] and the experimental side [(91.19 ± 3.13) cm2].IGF1:compared with the control side[(97.26 ± 1.02) cm2],the expression of IGF1 in the experimental side[(153.43 ± 1.18) cm2] was significantly higher (P < 0.05).IL-1 β:compared with the control side [(98.77 ±1.83) cm2],the expression of IL-1 β in the experimental side [(146.04 ± 1.24) cm2] was significantly higher (P < 0.05).IL-6:compared with the control side [(83.05 ± 1.22) cm2],the expression of IL-6 in the experimental side [(130.08 ± 1.03) cm2] was significantly higher (P < 0.05).TGF-β:compared with the control side[(85.10 ± 2.11) cm2],there was no significant difference between the experimental side [(84.04 ± 1.70) cm2,P >0.05].The expression levels of PI3K and AKT protein and mRNA were significantly higher than those in healthy controls (P < 0.05).Conclusion After the fracture of the condyle,the IGF1 was activated,and the activation of PI3K/Akt signaling pathway was activated,which resulted in the expression of IL-1 and IL-6.%目的 探讨在髁状突囊内骨折中骨形态发生蛋白(BMP)、成纤维细胞生长因子1(FGF1)、胰岛素样生长因子1(IGF1)、白细胞介素(IL)-1β、IL-6以及转化生长因子(TGF)-β的表现及其机制.方法 健康山羊15只,6~8个月龄,体重10 ~ 12 kg,雌雄不限,击打一侧颞颌关节区造成颞颌关节骨折,即刻进行CT扫描记录验证后的动物模型,取骨折处以及对侧健康处软骨组织.采用免疫组织化学法测定BMP、FGF1、IGF1、IL-1β、IL-6以及TGF-β,并分别检测两组磷酸肌醇3激酶(PI3K)、蛋白激酶B(Akt)的蛋白以及mRNA表达,进行对比分析.结果 BMP:对照侧为(101.51 ±2.16) cm2,与实验侧的(101.23±1.90) cm2比较,差异无统计学意义(P>0.05).FGF1:对照侧为(91.03±2.37) cm2,与实验侧的(91.19 ±3.13) cm2比较,差异无统计学意义(P>0.05).IGF1:实验侧为(153.43 ±1.18) cm2,与对照侧的(97.26±1.02) cm2比较,实验侧IGF1表达显著增高(P<0.05).IL-1β:实验侧为(146.04±1.24) cm2,与对照侧的(98.77±1.83) cm2比较,实验侧IL-1β表达显著升高(P<0.05).IL-6:实验侧为(130.08±1.03) cm2,与对照侧的(83.05±1.22) cm2比较,实验侧IL-6表达显著升高(P<0.05).TGF-β:实验侧为(84.04±1.70) cm2,与对照侧的(85.10 ±2.11) cm2比较,差异无统计学意义(P>0.05).实验侧PI3K和AKT蛋白以及mRNA表达水平明显高于对照侧(P<0.05).结论 髁状突囊内发生骨折后,IGF1首先被激活,进而使得其下游的PI3K/Akt信号通路激活,导致炎症相关因子IL-1和IL-6表达增强.
