SD大鼠光老化模型中氧化性损伤及灯盏花素的保护作用

         

摘要

Objective To investigate the mechanisms of breviscapin against skin photoaging and provide laboratory data for treatment of photoaging diseases. Methods Healthy SD rats were divided into 6 groups randomly and gavaged respectively. SD rats were irradiated to simulate skin photoageing by ultraviolet irradiation (UVA and UVB). Changes of the organization structure and collagen fiber of rat 9kin were observed. Changes of the contents of dermal hydroxyproline( HYP) and malond-ialdehyde ( MDA) , as well as the activity of super-oxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured. Results Significant photoaging-character in the rat of model group appeared in visual inspection. Oral administration of the model + medium-breviscapin 70 mg/kg Group ( E) and model + high breviscapin 140 mg/kg group (F) significantly prevented wrinkle formation in part of photoaging-character including skin erythema, desquamating, wrinkling and those photoaging characteristics were lighten compared with model group. In model + low-breviscapin 35 mg/kg group ( D), group (E) and group (F) : the photoaging characteristics gradually were lighten in Group E and F, the activity of SOD, GSH-Px and the content of HYP of dermal was decreased, while the content of the MDA was increased obviously (P < 0. 01). But there was no statistical difference between Group (E), Group (F) and between Group (B), Group (D) (P>0. 05). Conclusions Breviscapin might play anti-photoaging role by inhibiting the production of free radical and the lipid peroxidation, decreasing SOD, GSH-Px and MDA, elevating the activity SOD, GSH-Px and then reducing oxidative damage induced by ultraviolet irradiation.%目的 探讨灯盏花素抗光老化的作用机制,为防治光老化性疾病提供实验依据.方法 SD大鼠随机分成6组,模拟日光中UV( UVA+ UVB)长期照射,造成皮肤光老化模型.观察皮肤的组织结构和胶原纤维改变;测定各组SD大鼠皮肤中的超氧化物歧化酶(SOD)、谷胱甘肽氧化酶(GSH-Px)、丙二醛(MDA)、羟脯氨酸(HYP)含量.结果 灯盏花素中、高剂量组大鼠皮肤红斑、脱屑、皱纹等光老化改变明显比模型组轻.灯盏花素低、中、高剂量组皮肤光老化病理改变逐渐减轻.模型对照组SD大鼠皮肤组织中SOD、GSH-Px活性明显降低,HYP含量显著减少,MDA含量明显增加(P<0.01);灯盏花素中、高剂量组的皮肤组织中SOD、GSH-Px活性和HYP含量明显增高而MDA降低(P<0.01).结论 灯盏花素可通过抑制氧自由基的生成,降低SOD及GSH-Px消耗,提高SOD及GSH-Px活力,抑制MDA的合成和含量升高,从而减少紫外线引起的氧化损伤,发挥其抗光老化作用.
