三羟基异黄酮对转染APP695基因PC12细胞凋亡和功能的影响

         

摘要

Objective To explore the effect of Genistein (GST) on apoptosis and function of PC 12 cells transfected with APP695MT gene. Methods PC12 cells were transfected with pIRES2-EGFP empty vector and pIRES2-EGFP/APP695MT expression vector and were divided into control, APP695 transfected, GST intervention groups. Laser confocal microscopy was used to detect the apoptosis rates of PC12 cells, fluorescence analysis method was used to measure catecholamine (CA) secretory volume. Results Compared with control group, apoptosis rates of PC12 cells were higher and CA secretory volume was decreased in APP695 transfected group (P <0. 01). Compared with APP695 transfected group, apoptosis rates of PC12 cells were lower and CA secretory volume was increased in GST intervene group (P<0. 01). Conclusions GST can decrease the apoptosis rates of PC 12 cells inducing by APP695MT gene transfecting, increase CA secretory volume, protect transfected cells and improve the function of cells.%目的 以APP695MT基因转染PC12细胞为细胞模型,研究三羟基异黄酮(GST)对模型细胞凋亡和功能的影响.方法 用pIRES2-EGFP空载体和pIRES2 -EGFP/APP695 MT表达载体转染正常的PC12细胞,将细胞分为对照组、APP695转染组、GST干预组.应用激光共聚焦显微技术检测PC12细胞凋亡率,荧光分析法测定儿茶酚胺类(CA)分泌量.结果 APP695转染组与对照组比较,PC12细胞凋亡率明显升高(P<0.01),CA分泌量明显降低(P<0.01);GST干预组与APP695转染组比较细胞凋亡率降低(P<0.01),CA分泌量均明显增加(P<0.01).结论 GST可降低A PP695 MT基因转染引起的PC12细胞凋亡率,同时也可提高CA的分泌量,对转染细胞有一定的保护作用,并能改善其生理功能.
