白藜芦醇激活IGF-1R/AKT通路对退变椎间盘终板软骨细胞合成细胞外基质的影响及其机制

         

摘要

Objective To explore the effect of IGF-1R/AKT pathway on extracellular matrix synthesis of disc endplate chondrocytes stimulated by resveratroL Methods P2 generation of endplate chondrocytes were randomly divided into 5 groups and stimulated with corresponding reagent for 12 hours. Expression levels of IGF-1 R,pIGF-1R,AKT,pAKT,COLA II and aggrecan were detected by Western blotting technique. Results Resveratrol intervention could significantly increase the phosphorylation levels of both IGFR and AKT,and stimulate the synthesis of COLA II and aggrecan. AG1024 and SH-5 significantly inhibited the phosphorylation of both IGFR and AKT,and inhibit the synthesis of COLA II and aggrecan. Conclusions Resveratrol could stimulate the extracellular matrix synthesis of disc endplate chondrocytes via IGF-1 R/AKT regulatory pathway.%目的 探讨白藜芦醇对椎间盘软骨终板细胞合成细胞外基质的影响及其机制.方法 椎间盘软骨终板细胞培养,传代,P2代细胞培养48 h后,随机分为5组,各组加入对应试剂培养12h后终止培养,检测胰岛素样生长因子(IGF)-1R、pIGF-1R、蛋白激酶B(AKT)、pAKT、Ⅱ型胶原( COLAⅡ)、aggrecan的表达变化.结果 白藜芦醇干预后IGF-1R及AKT磷酸化水平显著增加,COLAⅡ、aggrecan基因转录水平上调,COLAⅡ、aggrecan合成增加;加入AG1024及SH-5抑制了IGF-1R及AKT磷酸化激活,COLAⅡ、aggrecan基因转录水平下降,COLAⅡ、aggrecan合成减少.结论 白藜芦醇可促进退变椎间盘软骨终板细胞合成细胞外基质COLAⅡ、aggrecan,抑制软骨终板退变,其机制与激活IGF-1 R/AKT信号通路有关.
