Objective To investigate the effects of decoction of yiqiwenyanghuoxue on Ang Ⅱ and aldosterone (ALD) in rats with early heart failure.Methods Model rats with chronic heart failure (CHF)were injected with Adriamycin and treated by decoction of yiqiwenyanghuoxue, then Ang Ⅱ and ALD levels were detected by radioimmunity.PPARγmRNA were detected by RT-PCR and protein by Western-blot Results Compared to the control group, Ang Ⅱ and ALD levels were significantly higher, but PPARγat mRNA and protein levels were significantly lower in CHF groups.Yiqiwenyanghuoxue decoction could improve the above changes.Conclusions Ang Ⅱ , ALD can be the predictors in early heart failure.Decoction of yiqiwenyanghuoxue can prevent myocardial remodeling and improve heart function through PPARγregulating Ang Ⅱ and ALD levels.%目的 探讨中药复方益气温阳活血方对早期心力衰竭大鼠血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)的作用机制.方法 以阿霉素尾静脉注射造模;超声检测评价其心功能、应用放射免疫法检测AngⅡ、ALD浓度、以RT-PCR法及Western印迹法检测过氧化物酶体增殖物激活受体γ(PPARγ) mRNA及蛋白质表达水平在益气温阳活血方治疗前后的变化.结果 随着心力衰竭的加重、心室重构的形成,模型大鼠AngⅡ、ALD含量进行性升高,而PPARγ mRNA与蛋白质的表达水平则进性降低.益气温阳活血方在心力衰竭的早期即可减少AngⅡ、ALD含量及上调PPARγ mRNA与蛋白质的表达水平.结论 AngⅡ、ALD可作为早期心力衰竭预警标志物;益气温阳活血方在心力衰竭早期可通过PPARγ信号通路调节AngⅡ、ALD的表达,防止心室重塑,改善心脏功能.
