RP-HPLC同时测定不同产地太白贝母中9种核苷类成分的含量

         

摘要

Objective To develop a sensitive and reliable RP-HPLC method for the simultaneous determination of nine nucleosides including uracil, cytidine, guanine, uridine, adenine, guanosine, thymidine, adenosine and 2'-deoxyadenosine from Fritillaria taipaiensis P. Y. Li that had been cultivated in different producing areas; To compare the contents of these nucleosides from different producing areas. Methods The analysis was performed on a Venusil MP C18 (2) column (4.6 mm×250 mm, 5 µm) with a gradient of methanol-water at a flow rate of 1.0 mL/min;the detective wavelength was set at 260 nm; the column temperature was set at 35 ℃. Results Uracil, cytidine, guanine, uridine, adenine, guanosine, thymidine, adenosine and 2'-deoxyadenosine were obtained in the good linear range of 0.269 5–16.17 µg/mL, 0.132 1–7.927 5 µg/mL, 0.095 5–5.73 µg/mL, 1.16–69.6 µg/mL, 0.48–28.8 µg/mL, 0.571 5–57.15 µg/mL, 0.526–52.6 µg/mL, 3.307 5–198.45 µg/mL, 0.530 5–31.83 µg/mL, respectively (r≥0.999 5);the recovery was in the range of 96.49%–101.65%(RSD≤2.92%). Conclusion The contents of the nine nucleosides from different producing areas have differences. Fritillaria taipaiensis P. Y. Li from Xianyi Village, Chengkou County, Chongqing City, whether the cultivated ones or wild ones, contain the highest level of nucleosides. The established method can provide references for the perfection of quality standard for Fritillaria taipaiensis P. Y. Li.%目的:建立反相高效液相色谱法同时测定不同产地太白贝母中9种核苷类成分(尿嘧啶、胞苷、鸟嘌呤、尿苷、腺嘌呤、鸟苷、胸苷、腺苷、2'-脱氧腺苷)含量的方法,并比较不同产地太白贝母中核苷类成分含量。方法采用Venusil MP C18(2)色谱柱(4.6 mm×250 mm,5µm),流动相为甲醇和水,梯度洗脱,流速1.0 mL/min,检测波长260 nm,柱温35℃。结果尿嘧啶、胞苷、鸟嘌呤、尿苷、腺嘌呤、鸟苷、胸苷、腺苷、2'-脱氧腺苷的线性范围分别为0.2695~16.17µg/mL、0.1321~7.9275µg/mL、0.0955~5.73µg/mL、1.16~69.6µg/mL、0.48~28.8µg/mL、0.5715~57.15µg/mL、0.526~52.6µg/mL、3.3075~198.45µg/mL、0.5305~31.83µg/mL,r≥0.9995;加样回收率为96.49%~101.65%(RSD≤2.92%)。结论不同产地太白贝母中9种核苷类成分含量存在差异,栽培品和多年生野生品均以重庆市城口县咸宜乡样品含量最高。本研究建立的方法可为太白贝母质量标准的完善提供依据。
