眼针疗法对大鼠脑缺血再灌注后海马组织脑源性神经营养因子表达的影响

         

摘要

Objective To investigate the effects of eye acupuncture on brain-derived neurotrophic factor expression (BDNF), and study its mechnism of cerebral protection from ischemia-reperfusion injury. Methods MCAO model rats established by suture method were randomly divided into the control group, the sham operation group, the model group and the eye-acupuncture group. After reperfusion 72 h, the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks. The expression of ischemic hippocampus BDNF mRNA was measured by RT-PCR, and the expression of ischemic hippocampus BDNF protein was detected by Western blot technology. Results After reperfusion 72 h, compared with the model group, the neurologic impairment marks of the eye-acupuncture group decreased obviously (P<0.05). The expressions of BDNF mRNA and protein in rat hippocampus after the eye acupuncture therapy were both obviously decreased (P<0.01). Conclusion Eye acupuncture therapy can increase the expression of BDNF in rat hippocampus with ischeniia-reperfusion injury, and protect brain by improving the repair of neurons.%目的 观察眼针疗法对大鼠脑缺血再灌注后海马组织脑源性神经营养因子(BDNF)表达的影响,探讨脑保护作用机制.方法 线栓法制备SD大鼠大脑中动脉梗死再灌注模型.将SD大鼠随机分为正常对照组、假手术组、模型组、眼针组4组.于再灌注72h后采用ZeaLonga评分法进行大鼠神经功能缺损评分,采用实时定量PCR方法检测缺血海马组织BDNF mRNA的表达,采用蛋白免疫印迹法检测缺血海马组织BDNF的表达.结果 缺血再灌注72 h后,眼针组大鼠神经功能缺损评分显著低于模型组(P<0.05),眼针组大鼠海马组织BDNF mRNA的表达和蛋白的表达较模型组均明显减少(P<0.01).结论 眼针疗法能提高大鼠缺血再灌注后海马组织BDNF表达水平,通过促进神经元的修复发挥脑保护的作用.
