siRNA沉默Nox1基因抑制胃癌细胞的生长

         

摘要

AIM: To investigate the inhibitory effect of synthetic NADPH oxidase 1 ( Noxl) short interfering RNA ( siRNA) on the growth of human gastric cancer SGC -7901 cell line. METHODS: The siRNA targeting Noxl mRNA was transfected into SGC -7901 cells and Noxl expression was determined by real - time PCR and Western blotting. Cell proliferation and apoptosis were examined by the methods of CCK -8 and flow cytometry, respectively. RESULTS: Noxl siRNA significantly inhibited the expression of Noxl in gastric cancer cells at both mRNA and protein levels. The apoptotic rate of SGC - 7901 cells transfected with siRNA was significantly higher than that of the control cells ( P < 0.01). The proliferation of SGC -7901 cells was also significantly suppressed in vitro (P<0.05) by siRNA transfection. CONCLUSION : Direct inhibition of Noxl expression by siRNA significantly suppresses the proliferation of SGC - 7901 cells in vitro, indicating a new approach of gene therapy for gastric cancer.%目的:探讨siRNA沉默NADPH氧化酶1(Nox1)基因表达对胃癌细胞的生长抑制作用.方法:合成Nox1 siRNA,采用实时定量PCR 和Western blotting 观察Nox1 siRNA 转染后胃癌SGC-7901细胞Nox1 mRNA及相应蛋白表达的变化;用CCK-8细胞增殖实验、流式细胞检测技术分别检测胃癌SGC-7901细胞增殖及凋亡的变化.结果:转染Nox1 siRNA 后的胃癌SGC-7901细胞Nox1 mRNA及蛋白表达明显受抑制.与对照组相比,干扰组SGC-7901细胞生长明显变缓(P<0.05),细胞凋亡率明显增高(P<0.01).结论:Nox1 siRNA 可明显下调靶基因Nox1的表达,在体外可抑制胃癌SGC-7901细胞的生长并促进其凋亡.
