黄金鲫源霍乱弧菌的分离鉴定及毒力检测

摘要

In order to identify the pathogen caused a serious mortality of cultural Carassius auratus anratus in Jiangsu province,the dominant bacterium stain YQ1 was isolated from the diseased fish and identified through bacterial phenotype characteristics,16S rRNA and gyrB gene sequence analysis.At the same time,these tests of artificial infection,virulence gene detection and antimicrobial sensitivity were carried ont.According to the phenotype characteristics,serotype characteristics and 16S rRNA and gyrB gene sequence analysis result,the isolate YQ1 was identified as non-O1on-O139 Vibrio cholerae.It contained lolB,toxR,hlyA Class,hlyA ET and ompU genes,but did not contain ctxA,ctxB,tcpA Class,tcpA ET,tcpⅠ,zot and stn/sto genes.The clinical signs of artificially infected Carassius auratus auratus with strain YQ1 were similar with the signs of naturally infected ones.The median lethal dose (LD50) in Carassius auratus auratus was 3.0×105 cfu.The antimicrobial sensitivity test showed that the pathogen was sensitive to Nalidixic acid,Enrofloxacine,Levofloxacin,Florfenicol,Tetracycline,rnDoxyeycline,Gentamicin,Streptomycin,Compound sulfamethoxazole,Cefotaxime,Furazolidone,Furadantin;intermediate sensitive to Neomycin;resistant to Sulfamonomethoxine,Ampicillin.The results provided non-O1on-O139 Vibrio cholerae strain YQ1 was a lethal pathogen.These results provided a basis for diagnosis and efficient control of the disease.%为研究引起江苏连云港某水产养殖场黄金鲫死亡的病原,本研究对黄金鲫内脏进行细菌分离、鉴定,并将获得的优势菌株进行动物回归实验、毒力因子检测和药物敏感试验.细菌的表型特征、16S rRNA和gyrB基因进化分析及血清型鉴定结果显示该菌株为非O1/非O139群霍乱弧菌(Vibrio cholerae),编号为YQ1.该菌株具有lolB、ompU、hlyA Class、hlyA ET、toxR等毒力基因,但不具有ctxA、ctxB、tcpA Class、tcpA ET、tcpⅠ、zot、stn/sto等毒力基因.动物回归实验显示该菌为致病菌,能够导致健康黄金鲫鱼死亡,其半数致死量LD50为3.0×105 cfu.该菌株对萘啶酸、恩诺沙星等12种抗生素敏感,对新霉素中度敏感,对磺胺间甲氧嘧啶、氨苄西林耐药.研究结果确定非O1/非O139群V.cholerae YQ1为引起黄金鲫大量死亡的致病菌.本研究为该病的诊断和防治提供实验依据.
