为研究猪链球菌2型(ss2)截短表达枯草杆菌素样蛋白酶(C5a-1)的免疫保护特性,本研究选择其B细胞线性表位优势区(aa 91～aa 323)进行PCR扩增,克隆至pET32a载体中进行原核表达,并接种小鼠鉴定其免疫保护性.Westemblot鉴定结果显示,截短表达的sspA重组蛋白分子量约45.6 ku,能够与SS2阳性血清反应,具有良好的反应原性.将纯化的重组蛋白经3次免疫CD-1小鼠,攻毒试验结果表明重组蛋白对小鼠的免疫保护率仅为菌体免疫所提供保护率的15％.本研究为利用该蛋白作为动物免疫制剂提供了实验数据.%For study the protection of trmcate subtilisin-like serine protease (sspA) against Streptococcus suis type 2 in mice,the truncate sspA encoding aa 91-aa 323 sequence was cloned and expressed in E.coli.The recombinant protein was about 45.6 ku and recognized by positive serum.In addition,mice was immunized with the purified protein and challenged with the fatal dose of S.suis 2.The results showed that the recombinant protein only provided 15％ protection in mice against the challenge with S.suis 2.
