黑美人粗肋草的组织培养研究

         

摘要

Taken the stem segments with side buds of Aglaonema commutatum Schott cv Treubii as explants,the effects of different sterilization methods,inoculation methods,and different concentrations of plant growth regulators in medium on in vitro rapid propagation of A.commutatum were studied.The results showed that:(1) the more suitable sterilization method for in vitro rapid propagation of A.commutatum was wiping the explants with 75% alcohol in combination with treating the explants with 0.1% HgCl2 for 20 min; (2) the more suitable inoculation method for adventitious buds induction was inoculating the explants into culture medium horizontally; (3) the most suitable culture medium for adventitious buds induction was MS+2.0 mg/L 6-BA+0.1 mg/L NAA,and under this culture medium the induction rate of 94.10% was achieved; (4) the more appropriate medium for multiplication was MS+4.0 mg/L 6-BA+0.05 mg/L NAA,and under this culture medium the multiplication coefficient of 6.30 was achieved and the most suitable generation numbers for proliferation culture was from five to six; (5) the more suitable culture medium for rooting culture was 1/2MS+0.2~0.3 mg/L NAA,and under this culture medium the rooting rate of 100% was achieved; (6) and the survival rate was higher when transplanting to peat or garden soil medium under temperature and humid preservation condition.%以黑美人粗肋草带腋芽的茎段为外植体,研究不同灭菌方法、不同接种方法以及在培养基中添加不同浓度的植物生长调节剂对黑美人粗肋草离体快繁的影响.结果表明:(1)采用75%酒精擦拭外植体和0.1% HgCl2灭菌处理20min较适合黑美人粗肋草的离体快繁;(2)采用芽眼朝上水平放置的接种方式较适合不定芽的诱导;(3)不定芽诱导最佳培养基为MS+2.0mg/L 6-BA+0.1mg/L NAA,不定芽诱导率达94.10%;(4)通过继代增殖培养筛选出较适合的培养基为MS+4.0mg/L 6-BA+0.05mg/L NAA,增殖倍数为6.30倍,最适继代代数为5~6代;(5)幼苗转入1/2MS+0.2~0.3mg/L NAA的培养基中生根效果较好,生根率达到100%;(6)移栽至泥炭或者菜园土的基质中并保温保湿,成活率较高,分别达到92.30%和88.00%.
