纸色谱法筛选知母中α-葡萄糖苷酶抑制剂

         

摘要

The enzymatic reaction method, paper chromatography and spectrophotometric method were combined to screenα-glucosidase inhibitors from Rhizoma anemarrhenaes in this experiment. The experimental conditions were as follows:the concentration of substrate was 10 mg/mL, amount of the enzyme was 100 µL,and the reaction time was 8 h in enzymatic reaction;the optimum condition of paper chromatography:volume of samp1e was 70 µL, open system was n-butyl alcohol-acetone-water(7∶2∶1), developing direction was radial direction;in spectrophotometric determination, after chromatography ,aniline phthalate was used as color developing reagent, then distilled water was used to elute, the wavelength for the absorbance reading was 365 nm.The experimental results show that aqueous extract, saponin, flavonoid and polysaccharide of rhizoma anemarrhenae, are good inhibitors of a-glucosidase,then Chinese herbal medicine rhizoma anemarrhenae is really excellent hypoglycemic plant resources.%  采用酶反应、纸色谱和比色相结合法筛选知母中α-葡萄糖苷酶抑制剂。实验条件:1)酶反应:底物浓度10 mg/mL、酶用量为100μL、反应时间8 h;2)纸色谱测定微量葡萄糖:点样量70μL、展开体系正丁醇-丙酮-水(2︰7︰1)、展开方向为径向;3)比色:色谱后苯胺-邻苯二甲酸盐显色,蒸馏水洗脱,比色波长为365 nm。实验结果表明知母水粗提物及其多糖、皂甙、黄酮组分均为α-萄糖苷酶抑制剂,而中草药知母确为优良的降糖植物资源。
