OBJECTIVE: To probe into the effects of three dosages of Aidi injection on the pharmacokinetic parameters of docetaxel in rats .METHODS: 24 SD rats were randomly divided into low dose group , medium dose group , high dose group and blank control group , with 6 SD rats in each .The low dose group , medium dose group and high dose group were respectively given tail intravenous injection of 5, 10 and 20 ml/kg of Aidi injection for 10 consecutive days , rats in blank control group received no process;all the four groups of rats were given same dose of docetaxel for 13.5 ml/kg at 10 min after the last administration at the 10th day.Each 0.4 ml blood was obtained from orbital venous plexus of four groups of rats at 2, 5, 10, 20, 30, 60, 120, 240, 480 and 600 min after the injection of docetaxel , and then was analyzed after pre-processing , the pharmacokinetic parameters were calculated according to the blood concentration .RESULTS:Compared with the blank control group , the plasma half-time ( t1/2 ) and apparent volume of distribution ( Vd ) of high dose group were significantly decreased; the t1/2 , area under curve of blood concentration-time and Vd of medium dose group were significantly decreased , the plasma clearance rate was significant increased, with statistically significant differences ( P <0.05 ).CONCLUSIONS: 10ml/kg of Aidi injection can accelerate the clearance of docetaxel in rats , which presented a strong induction .%目的:探讨三种剂量艾迪注射液对多西他赛在大鼠体内药动学参数的影响.方法:取24只SD大鼠随机分为艾迪注射液低、中及高剂量组和空白对照组,每组6只.艾迪注射液低、中及高剂量组大鼠分别尾静脉注射艾迪注射液5、10及20 ml/kg,连续给药10 d,空白对照组大鼠不做任何特殊处理;四组大鼠于第10日最后1次给药结束后10 min,尾静脉注射相同剂量的多西他赛13.5 mg/kg.四组大鼠于注射多西他赛后2、5、10、20、30、60、120、240、480和600 min于眼框静脉丛取血约0.4 ml,经预处理后进样分析,根据血药浓度计算药动学参数.结果:与空白对照组比较,艾迪注射液高剂量组大鼠血浆半衰期(t1/2)和表观分布容积(Vd)明显降低;艾迪注射液中剂量组大鼠t1/2、药时曲线下面积和Vd明显降低,血浆清除率明显升高,差异均有统计学意义(P<0.05).结论:10 ml/kg的艾迪注射液可加速大鼠体内多西他赛的清除,表现出较强的诱导作用.
