利用HPLC法对豆浆中的嘌呤物质进行检测分析,建立腺嘌呤、鸟嘌呤、黄嘌呤和次黄嘌呤4种嘌呤物质的多组分检测条件并进行方法评价.研究表明:利用Zorbax C18反相色谱柱(4.6 mm×250 mm,5.0 μm),流动相以KH2 PO4缓冲液(0.02 mol/L,pH 3.0),在流速1.0 mL/min,柱温25℃,进样量10 pL,检测波长(λ1)为254 nm的条件下,豆浆中的4种嘌呤达到最好分离,其精密度和回收率小于2%.%A feasible method using HPLC which was evaluated for analyzing Adenine,Guanine,Xanthine,Hypoxanthine from soybean milk was established in this paper.The chromatographic conditions were optimized as follows:Zorbax C18 column (4.6 mm ×250 mm,i.d.5.0 μm),254 nm by UV detector with 0.02 mol/L of KH2PO4 buffer solution (pH =3.0) as mobile phase,the flow rate was 1.0 mL/min,the column temperature was 25 ℃,and the sample size was 10 μL.The result showed that several purines can be separated effectively in these conditions with the precision and recovery under 2%.
