β-淀粉酶酶解产物含量的测定

         

摘要

Starch was hydrolyzed by heat-stable a-amylase,β-amylase.Their hydrolysates were determined by HPLC with differential refractometer detector.The hydrolysates of starch consisted mainly of maltose and maltotriose,trace amount glucose,and no sucrose.In the HPLC method,a good separation of hydrolysates was achieved.The determination of hydrolysates showed good reproducibility,and the RSD values of glucose,maltose and maltotriose were 2.08%,2.41% and 2.61%,respectively.The recoveries of glucose,maltose and maltotriose were 101.3%,98.4% and 93.5%,respectively.Among the 4 kinds of starch hydrolysates,that from the sweet potato starch by β-amylase was the lowest.%应用高效液相色谱示差折光检测法测定β-淀粉酶酶解淀粉产物中葡萄糖、麦芽糖和麦芽三糖的含量,出峰分离效果显著,重现性较好.葡萄糖、麦芽糖和麦芽三糖的RSD分别为2.08%,2.41%,2.61%.葡萄糖、麦芽糖和麦芽三糖加标后回收率分别在101.3%,98.4%,93.5%,回收率较高.4种植物淀粉酶解产物中,甘薯淀粉酶解产物中葡萄糖含量最低.
