脐橙皮中黄酮的超声波辅助双水相提取工艺研究

         

摘要

以低温烘干的脐橙皮为原料,利用超声波辅助乙醇-硫酸铵双水相体系提取脐橙皮中的黄酮。通过单因素试验和正交试验,考察了乙醇浓度、硫酸铵用量、超声波处理温度、超声波处理时间、液料比、回流提取温度、回流提取时间等因素对黄酮提取效果的影响。结果表明,最适宜的提取条件为:乙醇浓度40%,硫酸铵用量8 g,超声波处理温度50℃,超声波处理时间50 min,液料比25∶1(mL/g),回流提取温度60℃,回流提取时间3 h。在此条件下做了4次平行试验,平均得率可达8.173%,远高于相同条件下超声波非双水相法、双水相非超声波法和非超声波非双水相法,而且试验重现性好。%With low-temperature baked navel orange peel as raw materials, the flavonoid in navel orange peel was extracted by ultrasonic-assisted aqueous two-phase system of ethanol-(NH4)2SO4. The effect of ethanol concentration, amount of ammonium sulfate, ultrasonic treatment temperature, ultrasonic treatment time, the liquid-solid ratio, reflux extracting temperature, and reflux extracting time, on the extraction rate of the flavonoid, were studied by the single factor experiments and the orthogonal tests. The result showed that the most suitable extracting conditions were, the ethanol concentration of 40%, the amount of the ammonium sul-fate of 8 g, the ultrasonic treatment temperature of 50℃, the ultrasonic treatment time of 50 min, the liquid-solid ratio of 25∶1(mL/g), the reflux extracting temperature of 60℃, and the reflux extracting time of 3 h. Un-der the optimal conditions, four parallel experiments were carried out, and the average extraction rate could reach 8.173%, which was highly more than the yield by ultrasonic and no aqueous two-phase system, by aque-ous two-phase system and no ultrasonic, and by no ultrasonic and no aqueous two-phase system at the same conditions. The reproducibility of the experiments was good.
