中国被毛孢液体发酵培养条件优化

         

摘要

以中国被毛孢为试验菌株,对药瓶发酵培养基配方和发酵条件进行优化.在单因素试验的基础上,进行正交试验,结果表明最佳发酵培养基配方:葡萄糖3%,蚕蛹粉2%,土豆20%,KH2PO4 0.2%,MgSO4 0.1%,水解酪蛋白0.1%,复合维生素0.07%.最佳摇瓶发酵条件:初始pH 6.5,装液量100 mL/500 mL,接种量为5%,温度为16℃,摇床转速150 r/min.菌丝体干重为16.53 g/L,腺苷含量为1.85 mg/g.与优化前相比,菌丝体干重提高了36.2%,腺苷含量提高了46.8%.%This study took the Hirsutella sinensis as the experimental strain, and optimized its medium component of the shaking flask fermentation and ferment condition. The results of orthogonal experiment , which was based on the results of signal factor experiment , were as follows. The optimal medium component:glucose was 3 %, silkworm chrysalis powder was 2 %,potato was 20 %,KH2PO4 was 0.2 %, MgSO4 was 0.1 %, hydrolysis casein was 0.1 %,multivitamin was 0.07 %. The optimal fermentation condition: initial pH 6.5, liquid volume in flask was 100 mL/500 mL, inoculum concentration was 5 %, the temperature was 16 ℃, shaker's rotating speeds was 150 r/min. Based on this medium and ferment condition , the mycelium yield was 16.53 g/L and the content of adenosine was 1.85 mg/g. Compared to the original ferment condition , the mycelium yield and content of adenosine increased by 36.2%and 46.8%, respectively.
