不同固定剂及固定时间对三角帆蚌DNA提取效果比较

         

摘要

Freshwater pearl mussel, Hyriopsis cumingii, commonly called triangle mussel, belonging to family Unionin-dae, is mainly distributed in large lakes and rivers, and is rich in the five largest freshwater lakes in China; Poyanghu Lake, Dongtinghu Lake, Taihu Lake, Chaohu Lake and Hongzehu Lake. Extraction of genomic DNA from preserved specimen is a key step to study phylogeny and molecular genetics. This paper compared the effects of different preservatives on extraction of genomic DNA in a pearl-rearing shell, H. cumingii, collected from Jiangling, Jingzhou city, Hubei Province. Genomic DNA was well extracted by using the specimen fixed in 95% ethanol stored at 4℃, as well as in 100% ethanol at 4℃. The genomic DNA can be used as a template for PCR process of β-actin gene fragment.%以三角帆蚌(Hyriopsis cumingii)为对象,研究了无水乙醇,95%乙醇,4%多聚甲醛,福尔马林溶液,TE缓冲液5种固定剂不同固定时间对三角帆蚌斧足DNA提取质量及PCR扩增的影响.结果显示95%乙醇固定后提取的三角帆蚌基因组DNA降解少,DNA浓度高,OD_(260)/OD_(280)在1.8~2.0之间,OD_(260)/OD_(230)在1.5左右,表明95%乙醇固定后三角帆蚌基因组DNA的提取质量最好,再依次为无水乙醇、福尔马林溶液、4%多聚甲醛,DNA提取质量最差的是用TE固定的三角帆蚌.
