目的 研究广西人群Toll样受体4(TLR4)2244 G/A基因多态性与原发抗中性粒细胞胞桨抗体相关性小血管炎(AAV)的关联性.方法 采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法 检测122例AAV患者(AAV组)和225例健康成人(对照组)TLR4 2244 G/A位点的基因型,并收集实验对象 的临床资料和实验室资料.结果 (1)AAV组和对照组TLR4 2244 G/A位点基因型及等位基因分布频率比 较差异无统计学意义(P＞().05);(2)AAV组3种基因型间水肿发生率、尿素氮、肌酐、IgG水平相比较差异有 统计学意义(P＜0.05);(3)病理分析提示AAV组3种基因型间肾组织病理小血管炎活动指数及慢性指数评 分比较差异无统计学意义(P＞0.05).结论 广西人群中TLR4 2244 G/A基因多态性可能与水肿发生率、尿 素氮、肌酐、IgG升高具有关联性,可能与AAV的遗传易感性无关联性.%Objective To investigate the association of toll-like receptor ( TLR4 ) gene 2244 G/A polymorphism with primary anti-neutrophil cytoplasmic antibodies ( ANCA) associated small vasculitis ( AAV) in Guangxi population. Methods The genotype of TLR4 2244 G/A in 122 AAV patients ( AAV group) and 225 healthy adults ( control group) were analyzed by polymerase chain restricted fragments length polymorphism. The clinical data and laboratory data were collected. Results There was no significant difference in genotype distribution or allele frequency between the patient and the control groups ( P＞ 0. 05 ). The incidence of edema , the levels of serum urea nitrogen , serum creatinine and IgG were significantly different among the three genotypes of AAV patients ( P＜ 0. 05 ). No significant difference in AI or CI was observed among the three genotypes of AAV patients. Conclusion TLR4 2244 G/A gene polymorphism may be associated with the incidence of edema, the levels of serum urea nitrogen, serum creatinine and IgG in patients with AAV in Guangxi, but not be significant correlated with susceptibility to primary AAV.
