大鼠妊娠高血压胎盘脂代谢与脂肪酸结合蛋白4的相关性分析

         

摘要

目的 探讨胎盘组织脂代谢异常与脂肪酸结合蛋白4(FABP4)的关系在妊娠高血压(HDCP)发生中的作用.方法 SPF级雌性SD大鼠20只,随机分为妊娠对照组(PC组)和妊娠高血压组( HDCP组),每组10只,分别注射生理盐水、一氧化氮合酶抑制剂(L-NAME)50 mg/(kg· d)复制HDCP动物模型.测定大鼠血压、尿蛋白和胎盘组织HE染色.酶法检测胎盘中总胆固醇( TC)、游离胆固醇( FC)、胆固醇酯(CE)和游离脂肪酸(FFA)和三酰甘油(TG)的含量;运用qRT-PCR和Western blot检测胎盘中FABP4的表达,并分析其与TC和TG的相关性.结果 与PC组比较,HDCP组血压和尿蛋白水平均显著升高(P<0.05),胎盘HE染色显示HDCP组胎盘绒毛数目减少,结构不清;且胎盘组织中TC、FC、CE、FFA和TG含量及FABP4 表达较PC组显著升高(P<0.05);相关性分析发现TC、TG水平与FABP4表达呈正相关.结论 脂代谢异常可能在HDCP发病过程中起重要作用,而FABP4可能通过影响脂代谢,进而引起HDCP的发生与发展.%Objective To investigate the correlation between placenta tissue lipid metabolic disorder and lard type fatty acid binding protein 4 (FABP4) and its role in hypertension of pregnancy .Methods Twenty female SD rats (weight range from 250-300 g, SPF grade) were randomly divided into 2 groups (n=10 for each group), control group (Group PC) and hypertension of pregnancy group (Group HDCP).The rats in the 2 groups were injected with normal sa-line and nitric oxide synthase inhibitor (L-NAME) 50 mg/( kg· d) to induced hypertension of pregnancy model (HD-CP), respectively.Blood pressure and urine protein were recorded .Placental tissues were collected for HE staining .En-zymatic method was used to assess the total cholesterol (TC), free cholesterol (FC), cholesterol ester (CE), free fatty acid (FFA) and triglyceride (TG) content in placental tissues .qRT-PCR and Western blot were used to detected the expression of FABP4 in placental tissue.The correlation between the expression of FABP 4 protein and the contents of TC and TG in placenta tissues were analyzed .Results Compared with Group PC, the blood pressure and urine protein of Group HDCP were significantly elevated (P<0.05).HE staining showed that the decrease in the number of placental vil-li and unclear structure in Group HDCP .The contents of TC, FC, CE, FFA and TG and the expression of FABP4 were al-so significantly elevated in Group HDCP compared with Group PC (P<0.05).There were significantly positive correla-tions between TC or TG and FABP4 protein level.Conclusion Abnormal lipid metabolism may play an important role in the pathogenesis of HDCP .FABP4 may affect the lipid metabolism , which leads to the occurrence and development of HDCP.
