Objective To analyze the correlation between serotonin transporter promoter polymorphism (5-HTTLPR) and the susceptibility to chemotherapy-related fatigue in colorectal cancer.Methods The data of 5-HTTLPR genotypes and clinical data of 121 patients with colorectal cancer were retrospectively analyzed.The peripheral blood and clinicopathological data were collected.The correlations between 5-HTTLPR genotype and clinical pathological factors were analyzed by chi-square test.Unconditional logistic regression model was used to analyze the association between 5-HTTLPR variant and colorectal cancer chemotherapy-related fatigue.Results Chi-square test showed that chemotherapy-related fatigue was associated with 5-HTTLPR genotype (P=0.025).Unconditional logistic regression model showed that patients with genotype SS had a significantly higher risk of moderate to severe chemotherapy-related fatigue (OR=8.225, 95% CI 1.694~39.927, P=0.009) than genotype LL.Conclusion The genotype SS of 5-HTTLPR variant is an independent risk factor for moderate to severe chemotherapy-related fatigue in colorectal cancer.%目的 分析5-羟色胺转运体基因启动子多态(5-HTTLPR)长短变体(LL、LS、SS)与结直肠癌化疗相关性疲乏的易感性.方法 回顾性分析121例结直肠癌患者的5-HTTLPR基因分型及临床资料.收集患者外周血以及有关临床病理资料.2检验分析中到重度结直肠癌化疗相关性疲乏与5-HTTLPR基因分型及各临床病理因素的相关性,logistic多因素风险模型进一步分析5-HTTLPR长短变体对中到重度结直肠癌化疗相关性疲乏的独立影响.结果 化疗相关疲乏与5-HTTLPR基因型相关(P=0.025).Logistic多因素分析显示SS型基因组患者较LL型基因组患者发生中到重度化疗相关性疲乏风险明显增高(OR=8.225,95%CI 1.694~39.927,P=0.009).结论 5-HTTLPR长短变体SS型基因是发生结直肠癌中到重度化疗相关性疲乏的独立危险因素.
