GLS1基因在乳腺癌中的表达及在新辅助化疗中的意义

         

摘要

目的:探讨GLS1基因在乳腺癌中的表达及在新辅助化疗中的意义。方法收集134例乳腺癌患者为研究对象,入组患者经过穿刺活检确诊乳腺癌后,按照随机对照原则平均分成两组,每组67例。观察组在乳腺癌改良根治术前使用多西他赛+表柔比星+环磷酰胺( TEC )方案,进行3~4周期的化疗后再进行择期手术。对照组在改良根治术前不进行TEC化疗,直接手术摘除病变乳腺。术后两组患者采用免疫组化法,对病变组织中GLS1蛋白免疫组化染色。结果 GLS1基因在134例患者正常组织和乳腺癌组织中表达,但在乳癌组织中表达明显升高,差异有统计学意义( P<0.05);术前采用新辅助化疗后,癌组织中GLS1基因表达水平明显低于对照组,差异有统计学意义(P<0.05)。患者生存率与GLS1表达水平有关(P<0.05),表达水平越高生存率越低。结论GLS1基因在乳腺癌患者癌组织中高表达。在乳腺癌改良根治术前,使用辅助化疗可明显降低GLS1的表达水平;患者癌组织中GLS1基因表达水平与生存预后相关,GLS1基因在乳腺癌靶向治疗以及靶向治疗药物开发中可作为特异性的药物靶点之一。%Objective To investigate the expression of Glutaminase 1( GLS1) gene in breast cancer and its signif-icance in neoadjuvant therapy .Methods A total of 134 patients with biopsy proven breast cancer in Huizhou People′s Hospital were included.The participants were randomly divided into 2 groups (n=67).The participants in experimental group were treated with modified radical preoperative administration of docetaxel , epirubicin plus cyclophosphamide (TEC) scheme for 3~4 cycles of chemotherapy before elected surgery .In the control group, patients received no neoad-juvant chemotherapy before surgical removal of the breast .After the operation , the tumor from two groups were treated by immunohistochemistry .Results GLS1 gene was expressed both in normal tissues and breast cancer tissues , but it was sig-nificantly increased in breast cancer tissues (P<0.05).After the application of neoadjuvant chemotherapy , the expres-sion of GLS1 was significantly lower than that in the control group (P<0.05).The expression levels of GLS1 were divid-ed from 1 to 4, and the patients was followed up according to the expression level of GLS 1.It was found that survival rate was correlated with the expression level of GLS1 (P<0.05).The higher the expression level, the lower the survival rate was.Conclusion GLS1 gene is highly expressed in breast cancer .Before modified radical mastectomy of breast cancer , the administration of adjuvant chemotherapy can significantly reduce the GLS 1 expression levels .The expression level of GLS1 gene correlates with prognosis , and the GLS1 gene can be used as a target in treatment .
