目的 探讨黄芪对环孢素A诱导的肾病大鼠肾组织炎症反应及Snail的影响.方法 采用环孢素A制模,大鼠分为对照组、模型组和实验组.采用Masson染色观察肾间质改变,ELISA法检测TNF-α含量,免疫组化检测Snail 蛋白的表达及分布,RT-PCR检测Snail mRNA的表达水平.结果 (1)Masson染色:模型组肾组织胶原沉积显著加重,实验组较模型组胶原沉积显著减弱(P<0.05);(2)ELISA法:模型组TNF-α含量显著升高,实验组显著低于模型组(P<0.05);(3)Snail:①免疫组化:模型组表达较对照组显著增加(P<0.05),且多位于细胞核内,实验组Snail蛋白表达显著低于模型组(P<0.05).②RT-PCR:与对照组相比,模型组Snail mRNA的水平显著升高(P<0.05),实验组水平显著低于模型组(P<0.05).结论 黄芪能改善环孢素A诱导的间质炎症反应和抑制Snail表达.%Objective To evaluate the effects of astragalus mongholicus ( AM ) on inflammation and expression of snail in rats with cyclosporin A ( CsA ) - induced nephropathy. Methods Rats were randomly divided into three groups :control group, CsA group and AM group. Chronic: nephropathy was induced in rats with CsA. Renal interstitial fibrosis was graded according to Masson staining. TNF - β was quantifie.d with ELISA, while the expression of Snaill was assessed by immunohistocheruistry and RT - PCR. Results ( 1 )Prominent green collagen deposition was observed in CsA group ,while significant alleviation was revealed in AM group, according to Masson Staining ( P < 0. 05 ). ( 2 )Significant increase of TNF - α in CSA group, while significant reduction of TNF - α In AM group was revealed comparing with that in control group ( P < 0. 05 ). ( 3 )The protein and mRNA levels of Snail were both significantly increased in CSA group, while they were significantly reduced in AM group when comparing with those in control groups ( P < 0. 05 ). Conclusion Astragalus mongholicus attenuates interstitial inflammation and decreases the expression of snail in rats with cyclosporin A - induced nephropathy.
