从废次烟草中提取茄尼醇的工艺研究

         

摘要

[目的]探讨从废次烟草(根、茎、叶)中提取茄尼醇的最佳务件,从而降低茄尼醇的生产成本,以便工业化生产.[方法]通过单因素试验和正交试验,对茄尼醇的提取工艺条件进行了选择与优化.[结果]废次烟草样品最佳浸提条件为:浸提溶剂甲醇:正已烷为40∶ 60(V/V),浸提温度40℃,浸提时间3h,浸提液料比12∶1 ml/g.在最佳浸提条件下对废次烟草的根、茎、叶进行了提取和回收率试验,其提取率分别为0.06%、0.45% 、0.66%,其平均回收率(n=5)分别为89.6%、98.4%、102.5%.[结论]茄尼醇是一种弱极性物质,提取溶剂宜选弱极性物质,但考虑到废次烟草本身特点及提取成本,选择组合溶剂较理想且溶剂要适量;茄尼醇在高温下易氧化,故提取温度需温和.%[Objective] To discuss the optimum conditioas of extracting solanesol from the root, stem and leaf of discarded tobacco, so as to reduce the production cost and improve the industrialized production of solanesol. [ Method] The extraction conditions of solanesol were chosen and optimized by the single factor experiment and the orthogonal experiment. [ Result] The best conditions for solanesol extraction were using methanol/hexane = 40/60 ( V/V) as solvent at temperature of 40 ℃, 3 h of extraction, time and 12:1 ml/g of liquid to solid ratio, under the optimal experimental conditions, the extraction rate of solanesol were 0. 06% , 0. 45% and 0. 66% with the average recovery of 89. 6% , 98.4% and 102.5% in the root, stem and leaf of discarded tobacco respectively . [Conclusion] Solanesol is a weak polarity material, and its solvent is also with weak polarity. But in consideration of the characteristics of discarded tobacco and the extraction costs, the combination of solvents with appropriate ratio is better. Since solanesol is easily oxidized under high temperature, the extracting temperature should be moderate.
