土壤中纤维素降解真菌的筛选及酶活性分析

         

摘要

[Objective ] The research aimed to screen Fungi degradating wheat stem Cellulose and analyse the infect of C/N ratio on cellulose degradation enzyme activity. [Method] Using congo red appraisal medium method and filter paper degradation method screen Fungi from soil, characterizing the Fungi types by analysing fungi rDNA conservative domain sequence. The cellulose-enzyme activity of the two cellulose-decomposing strains cultured on different glucose and ( NH4)2SO4 content medium was analysised in this work. [Result]Two fungi strains exhibiting strong cellulolytic ability, named NY01 and NY02, were screened from soil in this study. Sequence analysing of fungi ITS conservative domain amplified from NY01 and NY02 DNA using the primer pair of ITS1 and ITS4 showed that NY01 and NY02 had 99% similarity in nu-cleotide sequence with Trichoderma and NY02 had 99% similarity in nucleotide sequence with Muco. Results show that the CMC enzyme activity and FPA enzyme activity was highest when the medium C/N ratio was 8:1. [Conclusion] The study provides the foundation for further exploration of the degradation of straw cellulose.%[目的]筛选降解小麦秸秆纤维素的真菌并分析C/N比对纤维素酶活力的影响.[方法]应用刚果红鉴定培养基和滤纸条降解度分析法筛选菌株,通过比对真菌rDNA的ITS区域序列鉴定菌株类型,通过调节培养基葡萄糖和(NH4 )2SO4的比例研究了纤维素降解酶活性.[结果]从土壤中筛选出有2株分解纤维素能力较强的真菌,分别命名为NY01和NY02;真菌ITS保守序列比时结果显示,NY01与木霉的相似性最高达99%,NY02与毛霉的相似性高达99%;培养基中C/N比值在8∶1时2个菌株的CMC和FPA酶活性均达到最高.[结论]该研究为进一步探索秸秆纤维素降解奠定了基础.
