透明质酸对绵羊卵母细胞体外成熟与早期胚胎发育的影响

         

摘要

[ Objective ] The research aimed to discuss the effects of hyaluronic acid ( HA ) at different concentrations on the in vitro maturation and early embryonic development of ovine oocytes. [ Method] With ovine oocytes as test materials,0, 1.5,3.0,4.5 mg/ml HA was added to the culture medium for the maturation in vitro and in vitro embryo of ovine oocytes to discuss its effects on the maturation in vitro and in vitro embryo of ovine oocytes. [ Result] When HA was 1.5 and 3.0 mg/ml, the maturation rate of ovine oocytes were 72.54% and 62.17% respectively, significantly higher than that of the control group (P<0. 05). When HA was 1.5 -3.0 mg/ml, HA have positive effects on maturation in vitro of ovine oocytes. When HA was 1.5 mg/ml, the cleavage rate and blastocyst rate were significantly higher than those in the control group and other added groups ( P < 0.05 ) and the total number of blastocysts cells was significantly higher than other groups ( P < 0. 05 ). These indicated that HA can promote the early embryonic development of ovine oocytes. [ Conclusion ] HA not only can promote the maturation of ovine oocytes,but it also play roles in the cell differentiation. HA at the concentration of 1.5 mg/ml was most favorable for the early embryonic development of ovine.%[目的]探讨不同浓度透明质酸对绵羊卵母细胞的体外成熟和早期胚胎发育效果的影响.[方法]以绵羊卵母细胞为试验材料,分别在绵羊卵母细胞体外成熟和体外胚胎培养液中添加0、1.5、3.0、4.5 mg/ml的透明质酸(HA),研究其对绵羊卵母细胞体外成熟和早期胚胎发育的影响.[结果]当HA浓度为1.5和3.0 mg,/ml时,绵羊卵母细胞的成熟率分别为72.54%和62.17%,显著高于对照(P<0.05).当HA浓度介于1.5~3.0 mg/ml时,HA对绵羊卵母细胞体外成熟有促进作用.当HA浓度为1.5 mg/ml时,绵羊卵母细胞的卵裂率和囊胚率均显著高于对照组与其他组(P<0.05),囊胚细胞总数显著高于其他各组(P<0.05).这说明HA可以促进绵羊卵母细胞的早期胚胎发育.[结论]HA不仅可以促进绵羊卵母细胞成熟,而且在细胞分化中也会起作用.当HA浓度为1.5 mg/ml时,最有利于绵羊的早期胚胎发育.
