华山松丛生芽增殖和生长研究

         

摘要

[Objective] The purpose was to supply references for breeding tissue-cultured seedlings of Pinus armandii Franch in large scale.[ Method ] The mature embryos of P. armandii were used as materials for inducing clustered buds and researching the influence of factors on the multiplication of clustered buds, such as medium, hormone levels, active carbon ( AC ) and picking off apical bud. [ Result ] DCR was the optimum medium for the multiplication and growth of clustered buds of P. Armandii; the multiplication of buds needed higher concn. of 6-BA and the addition of NAA went against the multiplication of clustered buds. AC showed obvious promoting effect on the elonggation of differentiated buds and its optimum addition amount was 2.0 mg/L. The multiplication and growth of the clustered buds in the medium of DCR +4 mg/L 6BA + 0.01 mg/L NAA + 30 g/L sucrose were better. Picking off apical buds could promote the multiplication of clustered buds, but their growth coefficient was smaller. [ Conclusion] The adventitious buds of P. armandiiwere induced successfully and its rapid multiplication was realized in this research.%[目的]为大规模繁育华山松组培苗提供参考.[方法]以华山松成熟胚为材料诱导丛生芽,研究培养基、激素水平、活性炭、摘除顶芽等因素对丛生芽增殖的影响.[结果]DCR是华山松丛生芽增殖和生长的最佳培养基,芽增殖需要较高浓度的6-BA;添加NAA不利于丛生芽增殖;活性炭(AC)对已分化芽的伸长有明显促进作用,且活性炭最佳添加量为2.0 mg/L;丛生芽在培养基DCR+4 mg/L6-BA+0.01 mg/L NAA+30 g/L蔗糖中增殖和生长状况较好;摘除顶芽可促进丛生芽增殖,但增殖系数较低.[结论]成功诱导出了华山松不定芽并实现了其快速增殖.
