氧化铝-活性炭吸附法纯化透明质酸的条件优化

         

摘要

[目的]研究用氧化铝-活性炭吸附法去除透明质酸(HA)中热原的优化条件,为生产高安全性的HA提供参考.[方法]通过分别对氧化铝添加量、第1次搅拌时间、活性炭添加量和第2次搅拌时间进行单因素试验和二次回归正交组合试验,以HA回收率和热原的含量作为评定指标,对氧化铝-活性炭吸附法纯化HA的条件进行了优化.[结果]优化后的条件:氧化铝添加量为80.00g/L,第1次搅拌时间为1.00 h,活性炭添加量为5.00g//L,第2次搅拌时间为2.00 h,此时的HA回收率最高,热原含量合格.[结论]对于发酵法制备的HA,用氧化铝和活性炭吸附的方法来去除热原比较简便,且处理后的HA热原含量合格.%[ Objective] The study aimed to research the optimized condition for eliminating the pyrogen in the hyaluronic acid( HA)with method of alumina and activated carbon absorption. [ Method] With the HA recovery and pyrogen content in HA as the evaluation index, the conditions for purifying HA with the method of alumina and activated carbon absorption were optimized through the single factor test and the square recurrence orthogonal test resp. with the 4 factors of Alumia dosage, first stirring time, activated carbon dosage and the second stirring time.[ Result ] The optimized conditions were as follows: the dosage of Alumia was 80.00 g, the first stirring time was 1.00 h, the dosage of activated carbon was 5.00 g and the second stirring time was 2.00 h, under which, the recovery rate of HA was the highest and the pyrogen content in HA was up to the standard. [ Conclusion] As for the preparation of HA with the fermentation method, it was relatively simple and convenient to remove the pyrogen with the method of alumina and the activated carbon adsorption, and the pyrogen content in HA after treatment could meet a criterion.
